Objective:To investigate the apoptosis-induced effect of disintegrin kistrin on rabbit lens epithelial cells (LECs)in vivo, and explore the relation apoptosis and the change of Cx43 in gap junction and typeIV collagen.Methods:12 New Zealand rabbits were randomly divided into two groups, and an extracapsular lens extraction(ECLE)was performed on the right eye of each rabbit. Their capsular bags of lens were injected with the concentration of 80ng·ml-1 of kistrin 0.2ml (the experimental group), and injected with the normal saline 0.2ml (the control group) just after the operation. All eyes were examined by slit-lamp microscopy at 1 day,3 day,5 day,7 day and 14 day after operation. The eyeballs were removed and then paraffin embedded at 14d.The apoptosis of LECs was observed by TUNEL staining and HE staining, and the expression of Cx43 was detected by indirect immunofluorescence. The expression of typeⅣcollagen was detected by immunohistochemical method and the collagen in capsular bag of lens was observed by MASSON staining.Results:There was no significant difference in the conditions of anterior segment between the two groups. Apoptosis index of the LECs in the experimental group which was (48.82±9.99)% was significantly higher than that in the control goup which was (12.36±7.53)%(P<0.01); The ratio of Cx43 positive fluorescent area in the experimental group which was (0.03±0.02)% reduce when compared with that in the control group which was (0.14±0.06)%(P<0.05); The gray value of typeⅣcollagen in posterior capsular in the experimental group is (136.38±2.69) and that in the control group is (120.53±6.53). The expression of typeⅣcollagen in the experimental group reduce when compared with that in the control group (P<0.05). There is statistically significant difference.Conclusion:The disintegrin kistrin can abate the expression of Cx43 between the LECs and the expression of typeⅣcollagen of posterior capsular, and induce apoptosis of LECs in vivo.
