Objective:To study the interaction between Raman spectrum of normal breast epithelial cell line(HBL-100)and carcinoma breast cell line(MCF-7);Raman spectrum as well as the change of biological tissue composition from normal tissues to malignant tissues of the breast. Characteristic Raman spectrum can be founded through the comparing and analyzing of the spectrum.Methods:Normal breast cell line(HBL-100)and carcinoma breast cell line(MCF-7) had been cultured,and the fresh specimens of breast cancer had been selected for this research after operation.Single cells suspension of normal and cancer tissues were made by mechanical dispersion method.Then the samples were detected by laser tweezers Raman spectrum technique.The supernates were collected backgroud spectrum by means of the same provocation power and exposition time.At the last,Single cells laser tweezers Raman spectrum were obtained by subtracting backgroud's from cell Raman spectrum.All experiments datas were handle with the oringin6.0 software,Single-cell Raman spectrums were analyzed by principal component analysis.Raman spectrum of normal breast cell line and carcinoma breast cell line were recorded,the result of which was analyzed by principal component analysis(PCA).Results:1. Through the study of Raman spectrum of the normal and carcinoma cells of the breast,We found that Significant differences of average Raman spectrums were founded between the normal cell and the carcinoma one ;The spectra line of carcinoma cell became strong at the whole; The strength of 936,1002,1298,1445 cm-1 increased; the peak of 1102 cm-1 shifted to 1094 cm-1 ;the peak of 484 cm-1 disappeared;Additionally,The structure and the amount of protein;nucleic acid;lipids and other such molecules had also changed.Average Raman spectrum of monocell was analyzed with the principal component of PCA, The result of which suggested normal cell and carcinoma cell could be differentiated with PCA,The discrimination rate was 87%(13/15).2. The average Raman spectrum had no manifest difference among every generation.3. The study of Raman spectrum of the normal and malignant tissues of the breast showed that: Significant differences of average Raman spectrums were founded between the normal tissue and the carcinoma tissue ; The spectra line of carcinoma tissue became strong at the whoe; The strength of 716,752,875,936,1002,1078,1152,1258,1296,1338,1445,1652 cm-1 increased ; The structure and the amount of protein;nucleic acid;lipids and other such molecules had also changed.Conclusions:1. Breast cancer cells and normal breast cells, compared to the Raman spectral lines change significantly.2. Breast cancer tissue and normal tissue compared to Raman spectral lines have also undergone a marked change.3. Judging normal breast cell and carcinoma cell with laser tweezers Raman spectrums is an efficient method.Single cells laser tweezers Raman spectrum technique can become a new way to diagnose cancer,which has very wide prospect.
