OBJECTIVETo investigate the expression of serum placental protein 13(PP13) and glycodelin in preeclampsia. To explore the relationship between PP13,glycedelin and preeclampsia. To explore the relationship between PP13 and glycedelin in preeclampsia and the relationship between PP13,glycedelin and gestation.METHODFourty-five women with preeclampsia (including 25 severe and 20 mild preeclampsia) and 40 normal pregnant women were recruited in department of obstetrics in Qilu hospital of Shandong University, according to the diagnostic code on the textbook "obstetric and gynecology".The two groups were matched in maternal age and gestational age.All cases had no other pregnant complications. Maternal peripheral venous blood 5ml was collected into test tubes for the isolation of serum.The levels of PP13 and glycedelin were detected by enzyme linked immunosorbent assay.Determine the optical density, using a microplate reader set to 450nm.Contruct a standard curve by plotting the mean obsorbance obtaine for each reference standard against its concentration, using the mean absorbance value for each sample, determine the corresponding concentration of PP13and glycodelin from the standard curve.The data was analyzed of two group T-test and Pearson relation were performed. RESULTS1.The levels of PP13 in serum of the preeclampsia group were 166.51±30.25 pg/mL while the levels of PP13 in serum of the controls were 61.83±17.63 pg/mL.The levels of PP13 in serum of the preeclampsia group were significantly higher than those of the controls (P<0.01).The levels of glycodelin in serum of the preeclampsia group were 112.79±24.79 pg/mL while the levels of glycodelin in serum of the controls were249.19±32.06 pg/mL.The levels of glycodelin in serum of the preeclampsia group were significantly lower than those of the controls (P<0.01).2. The levels of PP13 in serum of the mild preeclampsia group were 148.78±21.51 pg/mL while the levels of PP13 in serum of the severe preeclampsia group were 180.69±28.98 pg/mL. The levels of PP13 in serum of the mild preeclampsia group were lower than the severe preeclampsia group (P<0.01). The levels of glycodelinin serum of the mild preeclampsia group were 117.16±27.29 pg/mL while the levels of glycodelinin in serum of the severe preeclampsia group were 109.30±22.56 pg/mL.There are no remarkable differences between the mild preeclampsia group and the severe preeclampsia group (P>0.05).3.The coefficient correlation between PP13 and glycodelin in severe and mild preeclampsia were 0.187 and 0.216(P>0.05).So there were no statistical significance between PP13 and glycodelin.4.The coefficient correlation between PP13 in severe preeclampsia, mild preeclampsia,normal group and gestation were 0.746,0.744,0.724(P<0.05). So the levels of serum PP13 correlated positively with gestation. The coefficient correlation between glycodelin in severe preeclampsia,mild preeclampsia,normal group and gestation were0.141,0.110,-0.009(P>0.05).So the levels of serum glycodelin were not correlated with gestation.CONCLUSIONThe abnormal expression of PP13 and glycodelin may play a role in theoccurrence and development of the disease.They also play an important role in the trophoblast cell infiltration and promoting angiogenesis.But they have different mechanisms.They may be two different risk factors in the pathogenesis of preeclampsia.This experiment also confirmed that there is no correlation between the two factors.Further study of the relationship between PP13,glycodelin and preeclapsia may be possible to predict the future occurrence and development and may provide a new way of the clinical treatment.
