5-Aza-CdR On Ovarian Cancer Cell Growth And Gene Expression Of RASSF1A

[Objective]:To investigate the effect of 5-aza-2'deoxycytidine (5-Aza-CdR) on proliferation, methylation status and expression in mRNA and protein levels of RASSFIA gene in cultured human ovarian cancer cell line A2780 and OVCAR3.[Method]:treat ovarian cancer cell line A2780,OVCAR3 by different concentrations of 5-Aza--2'-Deoxycytidine (5-Aza-cdR), then detect the expression of mRNA and protein levels by reverse transcription-polymerase chain reaction (RT-PCR) and Western-blot (Western Blot); in the same time, ovarian cancer cell proliferation changes was observed by MTT detection, using methylation-specific polymerase chain reaction (MSP) test to dectect the methylation status of RASSFIA before and after drug treatment.[Results]:compared with control group,after drug treatment, when the 5-aza-2 'deoxycytidine concentration at 0.5,5,50umol/1, human ovarian cancer cells A2780, OVCAR3, could significantly inhibit tumor cell growth, and with 5-aza-2' deoxycytidine concentration increased, cell growth rate decrease; the control group cells, the RASSFIA mRNA and protein expression in A2780, OVCAR3 were seen less expressed; by the drug treated cells, RASSFIA mRNA expression is compared with the previous increase, and as the concentration increase the mRNA expression increased;RASSF1A gene promoter region showed DNA hyper methylation was found in A2780, OVCAR3,5-aza-2'deoxycytidine can reverse the Promoter methylation of RASSF1A gene.[Conclusion] 5-Aza-2'-deoxycytidine can change the tumor suppressor gene RASSF1A promoter methylation status of its re-expression, and thus inhibit ovarian cancer cell line A2780, OVCAR3 growth, and promote their apoptosis.
