Preliminary Study Of Abstract Of Periplaneta Americana To Prevent Experimental Liver Fibrosis Induced By CC1₄ In Mice

ObjectiveTo investigate the protective effect of the various doses of Abstract of Periplaneta Americana (APA) against liver fibrosis (LF) induced by CCl4 in mice.MethodsThe experimental LF model was induced by subcutaneous injection CCl4 in mice. APA (0.1,0.25,0.5 g·kg-1) or Colchicine (0.1 mg·kg-1) were administered intragastrically once a day for 6weeks. The hepatic function was evaluated by determining the levels of ALT, AST, ALB and TB in serum. And HA, LN, PC-ⅢandⅣ-C were determined by use of radioimmunoassay(RIA). The progression of hepatic fibrosis was confirmed by pathological analysis, grading of liver inflammation and staging of liver fibrosis (GS) were analyzed.ResultsSerum levels of ALT, AST, HA, LN, PC-ⅢandⅣ-C in the treatment groups were obviously lower than those in the model group, and the pathological results under microscope and degree of GS were much better as compared with the model group. The effect of high dose group was similar to the medium dose group and these two groups were much better than the low dose group.Conclusion1. The mice model of liver fibrosis induced by CCl4 is a simple,easy and cheaper experimental model. 2. APA can obviously cut down the indexes of ALT and AST in serums of the tested mice (P＜0.01). It is suggested that the drug probably has some influences on the protection of liver and decreasing the indexes of enzymes.3. APA can differently cut down the indexes of LN, HA, PC-ⅢandⅣ-C in serums of the tested mice (P＜0.01).It is indicated that the drug probably can improve collagen metabolism, decease the synthesis and (or) improve degradation of Extracellular Matrix (ECM), maybe has some effect of preventing Liver Fibrosis.4. APA has the different influence on reducing degeneration and necrosis of liver tissue. It is shown preliminary that the drug maybe has some effect on anti-hepatic fibrsis.5. The Experiment shown preliminary that The action of high dose group was similar to the medium dose group and these two groups were much better than the low group.
