ObjectiveThe purpose of study is to explore the expression of ERCC1 and TS in gastric cancer and adjacent tissues. With analysis of the relationships between the expression of ERCC1 and TS and clinical pathologic characteristic of gastric cancer, to provide the reference for the clinical individuation medication and the prognosis judgment. At the same time the correlation of expression of ERCC1 and TS was explored.Materials and MethodsThe expression of ERCC1 and TS were detected in 35 stomach carcinomatous tissues and 35 adjacent tissues by using immunohistochemistry SP method and RT-PCR technique. All of the tissues were collected from surgical procedures in our hospital from January 2008 to June 2008, which were confirmed by pathology and the data was intact. Expression of both gastric cancer and the clinicopathological characteristics and the relationship between the two expression in gastric cancer correlation statistical analysis.ResultsThe expression of ERCC1 and TS increased significantly(p<0.05) in gastric cancer tissues compared to the adjacent tissues; the over-expression of ERCC1 in gastric cancer was positively related to the differentiated degrees of the tissues (p<0.05), which means more ERCC1 were detected in highly intermediately differentiated adenocarcinoma than in those poorly differentiated ones, meanwhile it was not related to the patient's age, sex , the depth of invasion, lymphatic metastasis and clinical stages (p>0.05); the over-expression of TS in gastric cancer was obviously related to the depth of invasion , lymphatic metastasis and clinical stages (p<0.05). It indicated that the TS level is higher in the penetrated serosa(p<0.05), lymphatic metastasis(p<0.05) and Phase III-IV(p<0.05) than in that without invasion, without lymphatic metastasis and Phase I-II, respectively. However, it did not relate to the patient's age, sex and histological types (p>0.05). This implies the upregulating levels of TS is significantly correlated with some malignant behaviours of gastric cancer. Spearman rank correlation analysis system showed there is a positive correlation between the expression of ERCC1 and TS and gastric cancer.Conclusion1 The level of ERCC1's expression in carcinomatous tissues is higher than the level in the adjacent tissues. ERCC1 protein and mRNA expression associates with the degrees of differentiation which means the levels of ERCC1 protein and mRNA expression is higher in the advanced-differentiation adenocarcinoma than in the poorly differentiated one. However, it is not related with the patient's age, sex , the depth of invasion, clinical stages and lymphatic metastasis.2 The expression of TS in the stomach carcinomatous tissues is higher than in the tissues around the cancer. TS protein and mRNA expression are positively correlated to clinical stages, the depth of invasion and lymphatic metastasis, meanwhile, it has nothing to do with the patient's age, sex and the degrees of differentiation.3 The expression of ERCC1 and TS in gastric carcinomatous tissues is positively related.
