Objective: To investigate the association of acyl-CoA: cholesterol acyltransterase-2(ACAT-2)-734C/T polymorphisms with Hypertension and plasma lipids in Gansu Tibetans.Methods:To invesgate tibetans population in Gansu of age≥30 with methods of fradom sample.Body height and weight systolic blood pressure and diastolic blood pressure was recorded. The study covered 152 hypertensive and 128 normotensive controls in Gansu tibetans population. The segment of acyl-CoA:cholesterol acyltransterase-2 was amplified from DNA by polymerase chain reaction (PCR) . The PCR products were digested by rest riction endonuclease.Result: The frequencies of C and T in hypertensive group (0.796 and 0.204) were significantly different from those in the control group (0. 688 and 0.312,χ2=8. 652, P= 0.003) .The frequencies of -734C/T genotypes of CC , CT and TT wereO. 652,0. 289 and 0. 059 in hypertensive resp- ectively , and 0. 453,0. 469and 0.078 in conrols respectively. The genotypes of the variants were found to be in Hardy-Weinberg equilibrium in Tibetans groups. There was significant difference in genotype between hypertensive group and normotensive group (χ2 = 11.247 , P=0. 004)。The level of TC was higher in the subjects with CC genetype than that with CT and TT. But it was not significant difference(P>0.05).Conclusions:1.There is a polymorphism of ACAT-2-734C/T in Gansu Tibetans population (CC,CTandTT) .there are two allele(CandT).2.ACAT-2-734C/T polymorphism is associated with the hypertension.3. T allele may be a suspectibility gene of hypertension. 4. ACAT-2-734C/T polymorphism is not associated with the level of plasma lipids in Gansu tibetans population.5. ACAT-2-734C/T polymorphism is not associated with the BMI in Gansu Tibetans population.6.Age,overweight and dyslipidemia maybe the risk factor of hypertension in Gansu tibetans population.
