Objective:To research into the early microstructure changes of cut injury and electric burn of dog bile duct.Compare the difference of bile duct trauma caused by cut and electric burn injury.To explore the different mechanism of biliary restore between cut and electric burn bile duct injury and lay the theory basement for the reconstruction of the bile duct.Methods:A model of bile duct trauma was established in 35 native healthy half-bred dogs that were divided into contrast group,cut injury group and electric burn group randomly.The samples of the contrast group were get from the normal bile ducts of the five health dogs.Bile duct was excided by bistoury in cut injury group and electrotome in electric burn group respectively and was sutured with mucous membrane;Collect specimen on 1st week,2th week and 1st month after operation.Detect the expression of TGF-β1 and HIF-1a with immunohistochemical SP staining and observe the scene of tissue in hematoxylin eosin staining with light microscopy.Takes a quantitative analysis of positive-stained cells with image analysis system and analyze the result in statistics.Result:TGF-β1 and HIF-1a were observed in any tissue of normal bile duct and traumatic bile duct.TGF-β1 and HIF-1a were expressed highly in the tissue of model of bile duct trauma caused by cut injury and electric burn. TGF-β1 was expressed lightly and HIF-1a was not found in the normal bile duct.Statistics shows:TGF-β1 and HIF-1a were expressed higher in the cut injury group and electric burn group than contrast group(P<O.05).TGF-β1 was expressed higher in cut injury group than electric burn group contemporaneous (P<0.05).HIF-1a was expressed lower in injury group than electric burn group.The quantity of TGF-β1 increase gradually with the time and the counterpart of HIF-1a decrease on the contrary.Conclusions:The high expression TGF-β1 and HIF-1a of were related closely with the extracellular matrix deposition and scar proliferation in the healiing process of bile duct.The healiing process of bile duct trauma caused by electric burn may be longer than mechanical injury group.
