Objective: This study of cryosurgery avascular necrosis of the animal model, resection of the femoral head necrosis and the transfer of autologous greater trochanteric bone flap pedicled with blood vessels to reconstruct femoral head, in different time observing and analyzing the periost and aponeurosis of the greater trochanter bone flap surface, and other connective tissue fibers to invert the articular cartilage into law and transforming growth factor-β1 ( TGF-β1), in the process of cartilage metaplasia and the biological role to play。Methods: 8 to 12 months old Beagle 40, 7 to 10 kg body weight was used bilateral hindlimb autologous control, preparation cryosurgery bilateral avascular necrosis of the animals were randomly divided into two groups, each Group 20。The right femoral head after making model suture joint capsule, the right femoral head under different groups using different approaches: group A (bone flap treatment group): resection of the right femoral head necrosis frozen in liquid nitrogen after the transfer with the greater trochanteric bone flap pedicled with regional rotation Unit Cross-lateral vascular rotor to the femoral head and recycling; Group B (bone flap and rAAV2-TGF-61 gene therapy treatment group): The right femoral head with the greater trochanteric bone flap pedicled with blood vessels rotor femoral head recycling, intra-articular injection Expression of TGF-β1 of adenovirus vector; 3, 6, 12, 20, 32 two animals were killed four weeks each , a general observation on the bone , histopathology observation , immunohistochemistry and quantitative real-time fluorescence RT-PCR mRNA detection。Results: All animals right frozen tissue necrosis, and fiber bar coverage, debris-like tissue repair。Histologically section and I, type II collagen protein confirmed by immunohistochemistry that Group A the right of bone area six weeks since a hyaline cartilage cells, and each week increased, Group B the right bone flap district had in hyaline cartilage cells since three weeks, RT-PCR mRNA expression were detected with positive expression of the time-delay. Statistically P <0.05 with statistical significance. Conclusion:the periost and the aponeurosis of greater trochanteric bone flap able to metaplasia articular cartilage, TGF-β1gene expression, TGF-β1 is able to promot periosteum and aponeurosis of greater trochanteric bone flap to metaplasia articular cartilage, To supply base for the fiber surgical treatment avascular necrosis of the femoral head and the application of growth factor...
