Objective:The present study is designed to investigate the effect of Wenyang Huoxue recipe on TGF-bata/Smad Signal Transmission in preventing rats renal fibroblasts.Methods:Healthy male SD rats(n=32),weighing200g±20g,were divided randomly into four groups:the Sham-operated group(SOR),the Unilateral ureteral obstruction model group(UUO),the Wenyang Huoxie recipe group,the Monopril group.In addition to the sham-operated group rats,UUO model is used,that is,open the abdominal cavity of rats after anesthesia,and ligat the left ureter of the rats;the SOR rats had their left ureters manipulated but not ligated.Rats were administered orally from 24 hour after the obstruction,one time a day.At 14 day,blood was collected from abdominal aortic to prepare Serum.Blood creatinine and urea nitrogen were detected with automatic biochemical analyzer.HE staining was applied, renal tissues pathological change were examined by light microscop.Immunhistochemistry was applied to detect the expression of TGF-β1,Smad3 and Smad 7.Results:1.No clearly pathology change was seen in SOR group.Degeneration and vacuolation of tubular epithelium occurred and interstitial fibrosis with inflammatory cells infitltration were seen in UUO group.While the lesions in Monopril and Wenyang Huoxie recipe group were slighter than that of UUO group.2.There were no significant difference in the Serum creatinine of rats between all of groups(P>0.05).Compared with SOR,BUN increased significantly in UUO model group(P<0.05);Compared with UUO model group,BUN decreased significantly in Wenyang Huoxie recipe group and monopril group.3.The renel tissues expressions of TGF-β1 and Smad3 increased significantly,but the expression of Smad7 decreased significantly in UUO model group Compared with SOR(P<0.05);The renel tissues expressions of TGF-β1 and Smad3 decreased significantly and the expression of Smad7 increased significantly in Wenyang Huoxie recip and Monopril group,Compared with UUO model group(P<0.05),There were no significant difference in the expression of TGF-β1,Smad3 and Smad 7,between Monopril treatment group and Wenyang Huoxue recipe treatment group(P>0.05).Summary:Wenyang Huoxie recipe can prevente renal interstitial fibrosis by inhibiting the expression of TGF-β1 and Smad3,promoting the expression of Smad 7.
