Objective:1.To explore the relationship between the von Willebrand Factor(vWF) plasma levels and the attack of acute cerebral ischemic stroke(ACIS).2.To explore the relationship between the vWF gene Msp I polymorphism in intron 19 and ACIS.3. To demonstrate the allele frequency distribution of the vWF gene MspI polymorphism in intron 19 of the population in Kunming.4.To reveal the relationship between vWF gene Msp I polymorphism in intron 19 and its plasma levels.Methods:Specimens from 119 patients with acute cerebral ischemic stroke and 117 healthy controls were collected.And the plasma vWF levels were detected and its gene Msp I polymorphisms were analyst using polymerase chain reaction-restriction fragment length polymorphism method.Results:1.The plasma vWF levels of patients with ACIS were much higher than that of the controls(P<0.05).2.The gene frequencies of M+/M+,M+/M- and M-/M-genotype of the population in Kunming were 0.047,0.377 and 0.576,respectively. And the allele frequencies of M+ and M- were 0.235 and 0.765.The distribution of allele frequencies was different with that of Western population.3.The distribution of Msp I genotypes of patients with ACIS was quite different with that of the controls(P<0.05).The M+/M- genotype frequency was lower than that of the control group, while the M-/M- genotype frequency was higher,than that of the control group.4. Logistic regression analysis showed that elevated plasma vWF level,M-/M- genotype were the risk factors of acute ischemic stroke.5.The plasma vWF concentration of each genotype was significantly higher in the cerebral ischemistroke group compared with the control group.But the trends of these three genotypes in two groups were quite different.Conclusion:1.The elevated plasma vWF concentration contributed to the occurrence of ACIS.2.The frequencies distribution of vWF gene Msp I polymorphism in intron 19 of population of Kunming were different with that of western population.3.The vWF gene Msp I polymorphism in intron 19 had closely relationship with the occurrence of ACIS.The M-/M- was probably a genetic mark of the disease.4.The relationship between the vWF gene Msp I polymorphism in intron 19 and its elevtated plasma level was not clear by far.And it can not be confirmed that M-/M- genotype played a rolein the occurrence of ACIS by increasing the expression of vWF.
