Objective: To evaluate the expression of HMGB1, MMP-9, NF-κB and COX-2 in the disc degeneration process and further explore the relationship of HMGB1 and MMP-9, NF-κB and COX-2 in rabbit's lumbar disc degeneration .Method: 1. 24 adult Japanese white rabbits was used. And the way of destroying the posterior spinal stability was selected to establish animal models of disc degeneration. Groups: model group (L2-3 and L5-6) and self-control group (L3-4 and L4-5). 4 weeks, 8 weeks, 12 weeks after surgery We had X-ray,MRI examination to the two groups. Then killed the rabbits, observated the disc's morphous change with thrnaked eyes and histological change with HE staining, and tested the content of Collagen Type II and Aggrecan of disc. To make sure the disc degeneration and its level.2. To test the expression of HMGB1, NF-κB, MMP-9 and COX-2 of model and control group's discs in different time points with special antibody by immunohistochemisty SP.3. Based on immunohistochemical analysis of the results to further explore the various factors in disc degeneration in the meaning, as well as HMGB1, MMP-9 and NF-κB, COX-2 in lumbar disc degeneration in the relationship. Statistical analysed the results of immunohistochemisty SP, further explored the significance of above factors in disc degeneration, and the correlation of HMGB1 and MMP-9, NF-κB and COX-2 in disc degeneration.Result: 1. With time prolonging after surgery, the level of disc degeneration of the model group increased gradually. BUt the control group had no significant change .2. Immunohistochemistry, image analysis and statistical results showed that: in the process of disc degeneration, HMGB1 and MMP-9 had a positive correlation (r = 0.0497, P <0.05), NF-κB and COX-2 had a significantly positive correlation (r= 0.761, P<0.001).Conclusion:1. Posterior damage of spinal stability could establish animal model of disc degeneration.2. In the process of disc degeneration, HMGBI may up-regulate the expression of MMP-9, and enhance the degradation of matrix of disc.3. NF-κB may up-regulate the expression of COX-2 at the transcriptional level. In the process of disc degeneration, they might be involved in disc degeneration early inflammatory response.
