Objective To investigate the changes of the expressions of heat shock proteins 70 (HSP 70) and insulin and HSP70 protect effection on the gastric mucosa of rats with scald injury, and discuss the relationship between insulin and HSP70. Methods 55 healthy SD rats (provided by kunming Medical college) were set up scalded models,which were randomly divided into control group, scald group and scald+insulin group(treatment group). The gastric mucosa tissue sample separately after 3,6,12,24,48 hour were posted scald injury.In the treatment group, subcutaneous injection of 3u-kg-1 insulin after scald injury. the expression of HSP70 in the gastric mucosa was detected with immunohistochemical methods and analyzed by a micro-image analysis system, and at the same time we observed the pathological changes of the gastric mucosa tissue of each groups. Results In the scalded group, the expression of HSP70 was higher than the that in control group at the same time (P<0.05),6-12hr post scald injury the expression was increased,Following the expression of HSP70 decreased obviously 24hr post scald injury,48hr post scald injury the expression was increased, the expression of HSP70 in the treatment group was significantly higher than that the scalded group at most time points expel 12hr and at the same time (P<0.05), In the scalded group,the gastric mucosal injury index was higher than the that in control group at the same time (P<0.05),6-12hr post scald injury the gastric mucosal injury index was increased,24hr post scald injury the gastric mucosal injury index arrive higher, the gastric mucosal injury index in the scalded group was significantly higher than that the treatment group and at the same time (P<0.05),.In pathological observation, the control group manifested the intact gastric mucosal tissue formation, the scalded group showed obvious gastric mucosal tissue injury in the early phase of scald injury, while the treatment group showed much less severe injury than the scald group.Conclusions In the early phase of severe scald injury,the gastric mucosal tissue injury is very severe. It may be one of the vital mechanisms of insulin protecting the gastric mucosal tissue,which insulin increases the expression of HSP70 and decreases the gastric mucosal injury index in the gastric mucosal tissue of SD rats in the phase of scald injury.
