Objective To study the relationship of Leptin levels of bile and the differences in the form of Human benign biliary scar tissue. To explore the expression of Leptin, OB-R, IL-1β, and their relationship with scar formation in benign biliary scar tissue. To research the relationship of Changing trends between Leptin and IL-1, Leptin and its receptor in Human benign biliary scar tissue. To provide the experimental basis on prevention and treatment in Clinical benign biliary scar.Methods Being applied to chemical technologies Radioimmunoassay, Leptin levels were detected in the control group and benign biliary scar group. Using the Immunoenzyme staining (SABC) and the image analysis software to observe the expression of Leptin, OB-R and IL-1βrespectively in the control group and benign biliary scar group.Results Analysis of Leptin levels in the control group and benign biliary scar group using the Immunoenzyme staining showed that the normal bile contains Leptin protein and the value was 22.77±6.64, the measurements of Leptin in the benign biliary scar group was 61.18±17.68, significantly higher (P<0.01). The immunohistochemical results of scar group showed that the semi-quantitative integral values of Leptin, IL-1βand leptin receptor were 5.11±2.86,2.82±2.513,5.13±2.99 which significantly higher than other of the semi-quantitative integral value of 1.86±1.70,0.90±1.287,1.60±2.12 and statistically significant (P<0.01; P<0.05; P<0.01). Comparing with the expression of Leptin and IL-1βin the same group both showed a parallel trend. Comparison of the expression of Leptin and its receptor in the same group showed no significant correlation between changes.Conclusion Experimental results showed that there was a lot Leptin expressed in the normal human bile, which was about 5-6 times than that in the serum. Leptin levels in the bile of scar biliary was higher, indicating the correlation between Leptin and scar formation. The expression of Leptin, IL-1 and leptin receptor in the benign biliary scar tissue was positive, and they play a role in scar formation. Analysis of the expression trends of Leptin and IL-1βindicated that there was synergy between them which was an important factor to promote scar. Analysis of the expression trends of Leptin and its receptor indicated that the changes of the expression trends of Leptin receptor was not dependent on that of the Leptin, but was an independent factor which play a role in the process of scar formation.
