Periodontal diseases are those diseases that affect one or more of the periodontal tissues:alveolar bone,periodontal ligament,cementum and gingival.The incidence rate of the disease rose to 70%in China.Periodontal disease often results in loss of teeth, which causes dysmasesia and dyspepsia.Gingivitis,periodontal pocket formation, alveolar bone resorption and teeth loose are the four clinical symptoms of periodontal disease.Till now there are no ideal drugs and treatment to this disease.Alveolar bone resorption and formation of periodontal pocket are considered to be main symptoms which are the major reason of dysmasesia and dyspepsia.How to decrease bone resorption and promote bone modeling is the key subject for the therapy of periodontal disease.Traditional Chinese medicine herbs are thought as an important source of natural drugs.It has been widely used in the ancient China to treat musculoskeletal traumatic disorders.By the means of modern medicinal chemistry,Drynaria fortunei J.Smith, which virtual component is naringin,a type of flavone,was extracted and used in this research.Objective:To investigate the effects of Drynaria fortunei J.Smith naringin(DFSN) on proliferation and differentiation of human periodontal tissue cells.Methods:Human periodontal tissue cells were cultured as the model for experiment in vitro.Drynaria fortunei J.Smith naringin(DFSN) at different concentrations were used for treatment.Cell proliferation was determined by MTT method;cell differentiation was evaluated through the examination of alkaline phosphate(ALP) activities.Results:(1) Human periodontal tissue cells treated with 10μmol·L-1 and 1μmol·L-1 DFSN has obvious effects of promoting proliferation after 24 hours,0.1μmol·L-1 DFSN were also able to promote proliferation after 48 hours,and 0.01μmol·L-1 DFSN after 72 hours too(P<0.05).However,no significant changes in human periodontal tissue cells could be found when treated with DFSN at other concentrations in different culture time(P > 0.05).As the time goes by,except the 0.001μmol·L-1 group,DFSN in a concentration range of 10μmol·L-1 to 0.01μmol·L-1 showed a promoting effect on proliferation of human periodontal tissue cells.(2) Compare to control group,DFSN promoted ALP activities at the same dosages and point-in-time,similar to the results of MTT(P<0.05).10μmol·L-1 DFSN has stronger effects on increasing ALP activities of human periodontal tissue cells after 24h,1μmol·L-1 DFSN after 48h and 0.1μmol·L-1 DFSN after 72h too(P<0.01). It can not promotes the differentiation of human periodontal tissue cells at other concentrations for different culture time(P > 0.05).Along with time passing,ALP activities of human periodontal tissue cells increase except the 0.001μmol·L-1 group.Conclusion:Drynaria fortunei J,Smith naringin can promotes the proliferation and differentiation of human periodontal tissue cells.
