ObjectiveIn order to investigate the expressions of PTEN and Runx3 in chronic superficial gastritis, dysplasia and gastric cancer, and study the infection of Helicobacter pylori, to detect the relationship between the expression of PTEN, Runx3 and the Helicobacter pylori infection in gastric carcinoma, and to investigate their possible roles in the gastric carcinogenic mechanism, to provide evidence for prevention and therapy of gastric carcinoma as a therapeutic target.MethodsImmunohistochemistry S-P method was adopted to detect the expression levels of PTEN and Runx3 in 35 cases of chronic superficial gastritis,40 cases of atypical hyperplasia and 50 cases of gastric carcinoma; HE ,Warthin-Starry staining and rapid unease test method were used to detect Helicobacter pylori infection in the above-mentioned organizations. The data were analyzed by software SPSS 13.0, A difference was considered significance if the P value was below 0.05.Results1. Expression of PTEN:PTEN protein in patients with chronic superficial gastritis, dysphasia and gastric carcinoma were 94.3% (33/35), 80.0% (32/40) and 52.0% (26/50), PTEN protein in patients with chronic superficial gastritis group and the atypical hyperplasia group was significantly higher than expression in gastric carcinoma group, the difference has statistical significance (P<0.05), but chronic superficial gastritis and dysplasia group comparison of the difference was not significant (P>0.05), and gastric carcinoma pathology at the relationship between parameters, PTEN protein expression and the degree of differentiation of gastric cancer and lymph node metastasis has statistical significance (P <0.05), while patients with gastric cancer age, sex and size of gastric cancer, clinical stage (P>0.05);2. Expression of Runx3: Runx3 protein in chronic superficial gastritis, dysplasia and gastric carcinoma were 97.1% (34/35), 85.0% (34/40) and 56.0% (35/50), Runx3 protein in chronic superficial gastritis group and the atypical hyperplasia group was significantly higher than expression in gastric cancer group, the difference has statistical significance (P <0.05), but chronic superficial gastritis and dysplasia group compared, There was no significant difference (P>0.05), and gastric carcinoma pathology at the relationship between biology, Runx3 protein expression and the degree of differentiation of gastric carcinoma and lymph node metastasis has statistical significance (P <0.05), with the patient's age, sex and gastric size, clinical stage was no significant correlation (P>0.05);3. HP infection: Infection rates HP in patients with chronic superficial gastritis, dysplasia and gastric cancer tissue were 34.3% (12/35), 77.5% (31/40),and 70.0% (35/50). HP in atypical hyperplasia and gastric cancer tissues were higher than the prevalence of chronic superficial gastritis group rates (P<0.05), but not atypical hyperplasia and gastric cancer group comparison of HP infection rate was not significant, stomach HP infection in patients with age, sex and size of gastric carcinoma, clinical staging, differentiation degree, lymph node metastasis, such as no significant correlation (P>0.05);4. Relationship among PTEN, Runx3 and HP in Gastric cancer: by comparisons, correlation analysis was no significant difference (P>0.05).Conclusions1. The rates of PTEN, Runx3 expression in gastric carcinoma lower than the rate of which in superficial gastritis, dysplasia; suggesting gene of PTEN, runx3 at the occurrence of gastric carcinoma may be a disincentive;2. The rates of HP infection in dysplasia at gastric mucosa and gastric carcinoma tissue are higher than the rate of infection in chronic superficial gastritis mucosa, suggesting that HP infection had great importance during the process of progression from CSG, DYS to GC; 3. Through statistical analysis of compared gene PTEN, Runx3 and HP in gastric carcinoma occurrence and development of no significant correlation (P>0.05).MeaningTo study the pathogenesis of gastric carcinoma and a variety of carcinogenic factors in the occurrence and development of gastric carcinoma in the relationship between the provisions of strong evidence, its detection will improve the detection rate of early gastric carcinoma.
