The Clinic Curative Effect Observation Of Yi Qi Tong Luo Method Treating To PLGC And The Empirical Study To The Human Gastric Cancer In Orthotopic Implantational Nude Mice Model

Objective: To observe clinical curative effect on precancerous lesions of gastric cancer treated by YiQiTongLuo method, and to study its possible mechanism on regulating MMP-2 expression of human gastric cancer in orthotopic implantational nude mice model treated by the Yiqitongluo through animal experiment.Methods: Clinical part: case of the patients of precancerous lesions of gastric cancer were randomly allocated to treatment grope( 40 case) treated by YiQiTongLuo prescription, and control grope(40 case)treated by WeiFuChun, and observe the clinic effect after 3 months treatment. Experiment part: The experiment based on 60 nude mice models of orthotopic transplantation of human gastric cancer. They are divided randomly into six gropes(ten in each): celecoxib, rosiglitazone, HuangQi, Ezhu, HuangQi and Ezhu extract and blank controls. To observe the weight of orthotopic implantational tumor and depressant rate of orthotopic implantational tumor by the drug action after three and six weeks. The mRNA level of the expression of MMP-2 in orthotopic implantational nude mice model were determined by RT-PCR.Results: Clinical part: Clinical research showed that treatment group had symptomatic improvement in gastric abscess, painful abdominal, hypodynamia, anorexia, significantly better than control grope(P<0.05), there was no significant difference in diarrhea and gastric upset, (P>0.05). comparision of the total integral of symptoms had statistical significance(P<0.05)in YiQiTongLuo prescription group and control group spectively. the total effective rate in YiQiTongLuo prescription group was 82.5%, While the total effective rate of the control group was 65.0%, respectively, the difference had statistical significance(P<0.05). there was no significant difference in patho-atrophy(P>0.05), but there was significant difference in intestinal metaplasia and atypical hyperplasia(P<0.05). Experiment part: Sucessfully establish an orthotopic implantational model of gastric cancer in nude mice. The weight of orthotopic implantational tumor confirmed all medicine had manifest inhibit tumor growth, and tumor inhibition rate become more obviously following time. Tumor inhibition rate of HuangQi and Ezhu extract was obviously, comparing with those in control grope. RT-PCR showed that HuangQi and Ezhu extract could inhibit the mRNA level of MMP-2.Conclusion: YiQiTongLuo method was a effective method in the treatment of precancerous lesions of gastric cancer, with the probably antitumor mechanism was downregulating the expression of MMP-2.
