[Objective] To study the expression of P16 and Ezrin in cervical immature squamous metaplasia compared with 10 cases of normal cervical tissue, 10 cases of CINâ… , 10 cases of CINâ…¢and 10 cases of cervical squamous cell carcinoma and to find the expression regularity and effective markers combining the morphologic characters for diagnosis, in order to predict the prognosis of cervical immature mateplasia and adopt appropriate clinical managements. In order to research the role of human papilloma virus infection in the cervical immature squamous metaplasia development, we investigated four types of HPV infection conditions in those cases.[Methods] Immunohistochemistry was used to detect the expression of P16 and Ezrin in thirty cases of cervical immature squamous metaplasia compared with 10 cases of normal cervical tissue, 10 cases of CINâ… , 10 cases of CINâ…¢and 10 cases of cervical squamous cell carcinoma. Besides, DNA was extracted through the chosen samples fixed in 10% formalin and paraffin embedded. Four types of HPV infection conditions were detected by polymerase chain reaction (PCR) . All the data were analyzed by SPSS13. 0 software for windows.[Results] 1.The expression of P16 increased with the degree of severity (p<0. 01). The expression in CINâ…¢or SCC were statistically higher than that in CINâ… so as in normal tissues(P<0.01). The expression of P16 in CISM were statistically lower than that in CINâ…¢or SCC(P<0. 01). The difference between CISM and normal tissues had no statistical significance.2. The expression of Ezrin also increased with the degree of severity (p<0.01). There were statistical significances between SCC and CINâ…¢, as well as between CINâ…¢and CINâ… or normal tissues (P<0.01). The expression of Ezrin in CISM were statistically lower than that in CINâ…¢or SCC(P<0.01). The difference between CISM and normal tissues had no statistical significance.3. The infection rates of low risk HPV types in each group were similar. The infection rates of high risk HPV types in SCC and CINâ…¢were statistically higher than that in CINâ… so as in normal tissues (P<0.01). The difference between CISM and SCC or CINâ…¢had no statistical significance.4. There was no significant difference of high risk HPV infection rate between Ezrin positive and negative cases (P>0.05).[Conclusion] 1. The expression of P16 increase with the degree of severity and can be a supporting marker for distinguish diagnosis and clinical thereby guiding. It is supported that the CISM are benign based on the expression state of P16.2. The expression of Ezrin also increased with the degree of severity. The expression state of Ezrin in CISM also support that the CISM were benign.3. The high grade CIN and cervical squamous carcinoma have close relationship with high risk HPV infection. The development of CISM has no relationship with HPV infection.4. It is suggested that the expression of Ezrin can be heightened by high risk HPV infection.5. CISM is benign lesions. It is not the CIN. As the result of our study, cervical atypical immature squamous metaplasia (AIM) is different with CISM in etiology and pathogenesis.
