Objective:1.To establish the Bleomycin-induced pulmonary fibrosis model in rats;2.To observe the effects of Huayulifei prescription on the pulmonary fibrosis in rats;3.To study the effect of Huayulifei prescription on the contents of GSH,SODandMDA in the homogenate of the rat lungs;4.To study the differences of Huayulifei prescription and Hydrocortisone sodium succinate on pulmonary pathology,disparity of the content of GSH,SODandMDA in the homogenate of the rat lungs.Methods:Method:The eighty-four male wistar rats were randomly divided into model group, Huayulifei prescription group,hydrocortisone group and normal group.After being anesthtized with abdominal injection of pentobarbital sodium in a dose of 35mg.kg-1,the first three groups were then intratracheally dpipped with bleomycine(5mg.kg-1),while 0.9% saline in normal group.Treatments were started from the next day,the model and normal group were orally given 0.9%saline in a dose of 2ml,the Huayulifei prescription group was orally given Huayulifei prescription(17190mg.kg-1,2ml),ntraperitoneal hydrocortisone sodium succinate in a dose of 25mg.kg-1 in the hydrocortisone group.To observe the general condition of the rats:animal activity,type of figure,skin,mental tone,food and drink,excrement.7animals were randomly killed in each group at day7,14and28.Rats were intratracheally dpipped with bleomycine(5mg.kg-1),then were fixed lie on the back,cutting the stomach wall through the centerline of stomach blanket,rats were sentenced to death by exsanguinate the stomach aortic,and separating the lungs.Observing the lung appearance case of lungs,right lungs were putted into cold normal saline to remove blood,sucked dry by filter paper and cutted into small bricks,small bricks were made into 10%lung homogenate,lung homogenate was centrifugaled by 3000r.min-1 and the previous liquid was preserved in-70℃refrigerator to mensurateGSH,SODandMDA.Left lungs were isolated for Hematoxylin and eosin-staining.Results:1.General condition of rats in each group:eighty-four rats were involved in the analysis.There were no rats died in control group.,the action,body figure,nutrition and skin of rats were well in control group.The cough,dyspnea,decrement of activity,poor diet,thin physical statur,loss of weight and out of spirit of rats were found in model group.The rats'manifestations were similar to those in model group at early stage,but the situation turn better after being treat with Hydrocortisone except the activity and physical statur.The rats'in Huayulifei prescription manifestations were similar to those in model group at early stage,the rats'diet,physical stature,activity were better after treatment.2.General observation of lung:There were no abnormal findings of the rats in control group,whose lung profile was clear,pink,smooth and elastic.The dense bit haemorrhage,focal bleeding,lung volume augment were found in model group at day7,lung volume shrink,poor elastic,dense bit haemorrhage,each size dispersive tubercles were found and the lungs were off-white in model group at day14;The lung volum,lung hardness were more wore than before,more dispersive tubercles were found.The findings of lung morphology in Huayulifei group were similar to those in control group.The findings of lung morphology in hydrocortisone group were similar to those in model group.3.Lung coefficient:The lung coefficients of model group and hydrocortisone group were increased significantly than control group at day7,14and day28(p<0.01,p<0.05);the lung coefficients of huayulifei group were increased significantly than control group at day7,14(p<0.01,p<0.05),and the lung coefficients were similar to those in control group at day28,the lung coefficients of huayulifei group and hydrocortisone group were decreased significantly than model group(p<0.01,p<0.05);the lung coefficients of huayulifei group compare to that of hydrocortisone group were not statistically significant.4.Alveolitis and fibrosis:There were completely normal parenchymal structures in control group.The alveolar septums and pleura were thin,and the alveolar walls were formed in a fine pattern.The lung inflammation was very apparent at day7,there were moderate or severe alveolitis manifested by a thickening of alveolar walls accompanied by an accumulation of inflammatory cells within the parenchymal structures in model group at day 14.There were moderate or severe fibrosis manifested by a thickening of alveolar walls accompanied by an accumulation of fibroblast,collagen within the parenchymal structures in model group at day28.The inflammatory cells were decreased significantly in hydrocortisone group(p<0.01,p<0.05),the pulmonary fibrosis was decreased compared with that in model group,but not obviously at day28(p>0.05).The pulmonary fibrosis in Huayulifei group was alleviated significantly compared with those in model group(p<0.01), and was alleviated significantly compared with those in hydrocortisone groupat at day28(p<0.05).The alveolitis in Huayulifei group was alleviated significantly compared with those in model group(p<0.01,p<0.05).But the alveolitis and pulmonary fibrosis were significantly increased compared with those in normal group(p<0.01,p<0.05).5.The contents of GSH,SODandMDA in the homogenate of the rat lungs:The GSH in model group were lower than those in Huayulifei prescription group,hydrocortisone group and normal group at day7and14(p<0.01,p<0.05),but was gradually normal at day28;GSH were low in Huayulifei prescription and hydrocortisone group at day7,and were statistically significant compared with normal group,but were normal at day14.The SOD in model group were lower than those in Huayulifei prescription group,hydrocortisone group and normal group at day7(p<0.01),and were gradually high at day14,but also lower than those in other groups(p<0.01,p<0.05)and were also placed in low level at day28.SOD in Huayulifei prescription and hydrocortisone group were lower than those in normal group at day7(p<0.01)and gradually increased at day14,but were normal at day28.MDA in model group were higher than those in Huayulifei prescription group,hydrocortisone group and normal group at day7(p<0.01),and were gradually low at day 14,but also lower than those in other groups(p<0.01,p<0.05)but were also placed in high level at day28;MDA in Huayulifei prescription and hydrocortisone group were higher than those in model group at day7(p<0.01),but were lower than those in model group(p<0.01),MDA gradually decreased at day 14and were closely normal at day28.Conclusions:1.Huayulifei prescription and Hydrocortisone can decrease the lung coefficients of the Bleomycin-induced pulmonary fibrosis model in rats;2.Huayulifei prescription can ameliorate bleomycin-induced pulmonary fibrosis and alveolitis in rats;3.Huayulifei prescription can raise GSH,SOD content and decrease MDA content of lung homogenate,therefore can decrease the lung inflammation and suppress the development of pulmonary fibrosis induced by Bleomycin;4.Hydrocortisone can ameliorate bleomycin-induced lung inflammation and pulmonary fibrosis,but the result was not obvious.
