| Objective: By establishing rhesus uterine hemorrhage model by bacterial infection, we used compound components of sanchi to intervene the model. Then we wanted to find out matrix metalloproteinase -1,9and TIMP-1 express in rhesus endometrium were related to uterine hemorrhage by Bacterial infection. In the same time we wanted to discuss the mechanism of compound components of sanchi treating uterine abnormal bleeding. We wished all this could provid experiment evidence for clinical therapy of inflammatory menorrhagia.Methods: 11 cases of female adult rhesus with normal menstrual cycle were at random divided into three groups, including treating group(4 cases), model group(4 cases) and control group(3 cases). Injected the mixed bacteria to the two group(including the treating group and the model group) rhesus's uterus in the third menstruation day. The mixed bacteria were colibacillus, staphylococcus aureus and streptobacillus. Theirs proportion were 1:1:3. Five day's later, bacteria were cultured which indicated the bacteria were growing in their uterus. Then observed theirs menstruation change. Bred with compound components of sanchi to the treating group two days before the next menstrual cycle for seven days. Bred with the same 0.9%N.S to the model and control groups. After three menstrual cycles' therapy, we collected all rhesus endometrium specimens in the late excretion of the menstrual cycle and observed the changes of endometrium morphologa. The above-mentioned changes confirmed the animal models were succeeded. Then we used immunohistochemistry S-P to detect the MMP-1,9 and TIMP-1 expression in the three groups rhesus' endometriums .Result:â‘ we successfully established the rhesus uterine hemorrhage model by bacterial infection: From the results of evaluated strain ( colibacillus, staphylococcus aureus and streptobacillus were cultured in the uterus) ,the changes of rhesus menstruation and the result of observed morphology three ways, we could confirm the animal models were succeeded.â‘¡The results of MMP-1,9 and TIMP-1 express including: 1)MMP-1 expressed in the endometrial stroma, blood vessels, and glandular epithelium, but they expressed obviously in the stroma.2) MMP-9 mainly expressed in the endometrial glandular epithelium., and they expressed scatterly in the stroma and blood vessels,too.3) TIMP-1 expressed in the endometrial stroma, blood vessels, and glandular epithelium., expressed obviously vessels and around vessels. 4)Through imaging and statistics analyzing, we could get the below results: MMP-1 positive cells' area density and optical density in the treating group were below the model group, P<0.01, they had obviously statistics significance. Contrasted with normal group, P > 0.05 , no statistics significance. MMP-9 positive cells' optical density in the treating group was below the model group, P<0.01, they had obviously statistics significance. Contrasted with normal group, P>0.05, no statistics significance, but positive cells' area density in the three groups were not different, P>0.05, no statistics significance. TIMP-1 positive cells' area density and optical density in the three groups were similar, P>0.05, no statistics significance.Conclusions: All above suggested that rhesus uterine hemorrhage model by bacterial infection were successfully established. The injury of spiral artery were related to uterus abnormal bleeding by inflammation. Compound components of sanchi could resist inflammation and accelerate the repair of and contraction the spiral artery, therefore they could achieve hemostatic purpose. The over-express of MMP-1,9 lead to the unbalance of MMPs and TIMPs. The over-express of MMPs perhaps was a key factor of uterus abnormal bleeding, which directed MMPs took part in the process of lesion endometritis. Compound components of sanchi perhaps inhibited the over-express of MMP-1,9 so could regulate the balance of MMPs and TIMPs, which brung into the role of treating uterus abnormal bleeding. |
