The Research On The Mechanism Of Inhibiting Gastric Juice Secretion To Ulcer Rats

Objective:To explore the action mechanism of Shuganhewei Pill on inhibiting the secretion of gastric hydrochloric acid and pepsin.Method:60 rats,famale and male are half and half,weighing 180-200g,were divided into 6 groups:Shuganhewei Pill groups respectively at low,moderate and high doses,model group, Weisu Powder control group,and blank group.Each group contains 10 rats.Shuganhewei Pills with 2.18g/kg,1.09g/kg, 0.54g/kg were respectively administrated to high dosage, moderate dosage and low dosage groups.Weisu Powder with 1.52g/kg was administrated to Weisu Powder group.An equal volume of physiological saline was administrated to model group, dissolved into distilled water separately.Every group was instilled with the medicine about 2ml a time,7 days continuously.After the end instillation,the rats were anaesthetized with chloral hydrate,Cut the hair of the belly, routine sterilizate.Cut a notch about 2.5cm long along the belly white line under the xiphoid.Take out the stomach carefully,ligate the joints between pylorus and duodenum, injecte every medicine from the duodenum immediately.Sew up the wound,sterilize,send the rats back to the cage,restraint the intaking of food and water after the operation.Take off the cervical vertebra to put to death after 24h,take out the stomach,collect the gastric juice,centrifuge for 15min at 3000 rpm,extract upper heat removing,It is the total gastric juice amount.Cut off along the greater curvature,observe stomach mucous membrane damage situation,calculate stomach mucous membrane damage index according to GUTH method.Results:1.Compared with the blank group,Weisu powder group,the ulcer numbers in the model group were visibly increasing,which is of significant difference(P＜0.05), stomach mucous membrane necrosis was more obvious.The ulcer numbers in the Shuganhewei Pill groups was much lower than the model group,which is of apparent difference(P＜0.05),stomach mucous membrane has not obviously necrosised.This indicates that Shuganbewei Pill has obvious protective action on the stomach mucous membrane.2.Shuganhewei Pill has prominent function of inhibiting the secretion of gastric hydrochloric acid and pepsin(P＜0.01), high,moderate,low dosage group have result this,and have obvious dose-effect relationship.High,moderate dosage group are superior to positve control group(P＜0.01).3.Shuganhewei Pill can promote D cell hyperplasia,inpel G cell wither(P＜0.01),high,moderate,low dosage group have result this.High,moderate dosage group are superior to positve control group(P＜0.01).Conclusion:Shuganhewei Pill has prominent function of inhibiting the secretion of gastric hydrochloric acid and pepsin;Shuganhewei Pill has obvious protective action on the stomach mucous membrane;Shuganhewei Pill can promote D cell hyperplasia,inpel G cell wither.
