Objiect: ulcerative colitis (UC) is belonging to the category of "recurrent dysentery"and"diarrhea"in Traditional Chinese Medicine (TCM).the pathogenesis of UC is toxin chokeing the large intestine, entrails disequilibrium, injury the system of intestine.clinical manifestation is abdominal pain, diarrhea and bloody stools, contemporary study show that, once UC comes out,the immunity of mucous membrane of intestinal tract disharmony, and the immunity of all over the body mucous membrane disharmony, the cytokine of IFN-γpromoted inflammation up-regulation,that can induceed and promoted the inflammation; the cytokine of IL-4 promoted inflammation down regulation,that can induce the effect of antiinflammatory insufficient.and those study shows that it have direct correlation between the expression and externalization of the cytokine and the extent of inflammation of UC.the objiective of this experiment is to investigate the effect of Sanpi decotion in the weight of mice spleen , pathological changes of colon,and the IL-4 and IFN-γof blood serum and spleen in UC. To investigate the therapeutic effect of sanpi decotion in ulcerative colitis(UC) mice and discuss its therapeutic mechanism.Methods:40 mice were randomly divided into four groups including Normal group, Model group, Sanpi decotion group, SASP group,each group 10 mice, in accordance with Heller et used method that erect model of the UC of mice by QXZ. Sanpi decotion group and SASP group were respectively administered at Sanpi decotion(0.55 g.ml-1) and SASP (0.05 g.ml-1) to intragastric administration. Every day caculated on Disease activity index(DAI).At 10th day to execute all mice , caculate on pathological changes of colon, to investigate the weight changes of spleen,and to utilize ELISA method to investigate the IL-4 and IFN-γof blood serum and spleen in each group.Results: 1. general state of health and caculate on DAI: Model group, Sanpi decotion group, SASP group,each group colociysis after 24 hour comes out weight lessen, diarrhea, lazy act, extrados, anorexy; all the Model group mice appearance Loose stools,5 mice comes to bloody stools, compare to Normal group having significant difference in DAI. Sanpi decotion group and SASP group the symptom start to relief at fourth day, the weigt gradually increase, compare to Model group having significant difference in DAI;at tenth day , Sanpi decotion group and SASP group the symptom start to relief significantly, spirit turns improved, appetite increased, no bloody stools,sometimes Loose stools,and the Sanpi decotion group mice symptom' amelioration better than SASP group,the two groups not have significant difference.2. injury of tissue: The mice intestinal tract of model group can be seen obvious congestive and edema, vertical cut along the mesentery can be seen scattered in the intestinal wall mucosa and erosion of the ulcer, lesions mainly in the distal colon to the rectum the most serious diseases. The intestinal tract of mucous membrane of Sanpi decotion group and SASP group were significantly better than the model. Under light microscope,the pathological form: the model group mice intestinal erosion and ulcers can be seen the formation of lesions confined mainly to the mucosa and submucosa, the lesion has a large number of neutrophils and lymphocytes infiltration; histological score was significantly higher than the normal group, Sanpi decotion group and SASP group; The mice of Sanpi decotion group significant improvement in colonic lesions, mucosal erosion and ulcer-free, and only a small amount of inflammatory cell infiltration; SASP groups of mice had no obvious intestinal erosion and ulcers still there is inflammatory cell infiltration.3. The condition of Spleen weight: the model group of spleen weight in mice significantly reduced compared with the normal group were significantly different (P <0.01); Sanpi decotion group and SASP group soup spleen weight than the model group were significantly increased.4. Each group of serum IFN-γ, IL-4 content: the model group, serum IFN-γwere significantly increased, IL-4 were significantly lower compared with the normal group were significantly different (P <0.01), Sanpi decotion group and SASP group content of IFN-γcompared with the model group were significantly lower and there is statistical significance (P <0.01), IL-4 content compared with the model group were increased, Sanpi decotion group and the Model group There was a significant group (P <0.01), Sanpi decotion group and SASP group comparison had no significant inter-group differences.5. Each group of spleen IFN-γ, IL-4 content: the model group, serum IFN-γwere significantly increased, IL-4 were significantly lower compared with the normal group were significantly different (P <0.01), Sanpi decotion group and SASP group content of IFN-γcompared with the model group were significantly lower and there is statistical significance (P <0.01), IL-4 content compared with the model group were increased, Sanpi decotion group and the model group were statistically significant (P <0.01), Sanpi decotion group and SASP group comparison had no significant inter-group differences.Conclusion: The Sanpi decotion group has the exact role of the improvement on animal model of UC, presume its mechanism may be by regulating the Thl/Th2 balance and improve immune function, promote healing UC.
