Aim1 .To established the two dimensional polyacrylamide gel electrophoresis (2-DE) expression maps of total protein in PBMC of the Liver and Gallbladder Wetness-heat Syndrome of Chronic Hepatitis B and the normal persons and analysis the different expression proteins. According to the analysis and comparison of the differences among the 3 group' proteomics express,we study the different proteins in the PBMC of the Liver and Gallbladder Wetness-heat Syndrome of Chronic Hepatitis B,which can be helpful to provide mechanisms for approaching the nature of Liver and Gallbladder Wetness-heat Syndrome and CHB.2. To investigate the Pharmacological mechanisms of the qingganlishi Decoction on patients with Chronic Hepatitis B (CHB),proteomic methods were used to find and identify differential expression proteins in the peripheral blood mononuclear cells (PBMC) of patients with CHB, who were treated by the qingganlishi Decoction.Methods:1. Forty patients were randomly divided into2groups.Each patient in the 2 groups was treated with Qingganlishi decoction or western medicine lamivudine. The clinical effects were compared after 3-month treatment.2. The PBMC of the Liver and Gallbladder Wetness-heat Syndrome of Chronic Hepatitis B and the normal persons were isolated by using Lymphocytes separation medium. 2-DE was applied to separate the total proteins of the three groups to gain different expression proteins and establish 2-DE image and analysis them by PD-Quest 7.3.1 software. Different expression proteins were identified by MALDI-TOF-MS . MSDB database searching was used to identify proteins. In order to measure peptide mass fingerprinting,MSDB and Swiss-Prot protein database were retrieved to identify different expression protein. Results:1. As compared with western medicine group,Qingganlishi decoction showed no significant difference in therapeutic effects (.P>0.05), but the improvement in symptoms and the Liver and Gallbladder Wetness-heat Syndrome seems to be significant in Qingganlishi decoction group (P<0.05).2. The expression maps of proteomics in PBMC of the Liver and Gallbladder Wetness-heat Syndrome of Chronic Hepatitis B and normal persons were established .The average spots' number of PBMC is (980±280) in healthy person, (960±21) in the Liver and Gallbladder Wetness-heat Syndrome of Chronic Hepatitis B patients and (997±15) in this patients after treated by the Qingganlishi Decoction. We match the spots of 3 gels refer to the gel of the healthy people group.The average matching spots of PDGYHFs is (816±65) in healthy person,(844±87)in the Liver and Gallbladder Wetness-heat Syndrome of Chronic Hepatitis B patients,and(817±55) in this patients after treated by the qingganlishi Decoction..The average matching rate is 85%,88%and 82%.3. We selected 19 spots with different expression to make mass-spectrum identification.The identified proteins include cytoskeleton proteins,metabolic enzymes,signal transmission proteins, intoxication proteins blood proteins and so on.Conclusion:1. The expression maps of proteomic including in the Liver and Gallbladder Wetness-heat Syndrome of Chronic Hepatitis B patients and normal persons were established.2. These different expression proteins between the untreated CHB and after treated by the Qingganlishi Decoction people,may play important roles in biological and clinical effects between the treated and untreated.
