| Objective:To observe whether the selective COX-2 inhibitor celecoxib can inhibit the proliferation of malignant lymphoma Raji cells,to investigate the synergistic effects and possible mechanism of adriamycin combined with celecoxib on Raji cells.Methods:1.The Raji cells were treated with celecoxib and adriamycin in different concentration respectively,then the growth inhibiting effects were observed by MTT assay.2. The Raji cells were treated with invariable adriamycin combined with celecoxib in different concentration. The growth inhibiting effects were observed by MTT assay in order to assess the interaction.3 .The effects and probable molecular mechanism of the different treatments (celecoxib,adriamycin,celecoxib+adriamycin) on Raji cells were analyzed by flow cytometry,Hoechest33258 fluorescein stain, cas-pase-3 activation assay and RT-PCR.Results:In certain range of concentration,celecoxib could obviously inhibit proliferation of Raji cells in a dose- and time- dependent manner(P< 0.05).The growth inhibition present synergistic effects,that induced by 0.5mg/L adriamycin combined with 7.5-20mg/L celecoxib,while the inhibition present addition effects which induced by 0.5mg/L adriamycin combined with 5mg/L celecoxib. The effects that 0.5mg/L adriamycin combined with 7.5mg/L celecoxib induced Raji cells apoptosis and caspase-3 activation was more prominent than they did respectively(P< 0.05).Celecoxib could down-regulate the level of survivin mRNA,and the effect was more obvious while it combined with adriamycin.Conclusion:1. In certain range of concentration,celecoxib could obviously inhibit proliferation of Raji cells in a dose- and time- dependent manner.2. In certain range of concentration,the growth inhibition induced by adriamycin combined with celecoxib present synergistic effects.3. Celecoxib could enhance the anti-Raji cells effects of adriamycin, and the molecular mechanism may relate to activation of caspase-3 and down-regulation of survivin. |
