Objective1、Investigate the influence of combined application of bicyclol and adriamycin to the apoptosis of human lymphoma Raji cells.2、Investigate whether the bicyclol, which is a kind of liver protective agent, has protective effect to the lymphoma cells damage process cause by adriamycin. This will provide clinical reference for optimal combined application of chemotherapy drugs and liver protection drugs.MethodObserve the impact of separate application of bicyclol or adriamycin and combined application on human lymphoma Raji cells. Study the cell apoptosis employing MTT colorimetry method, flow cytometry and Western blotting. Investigate the impact of bicyclol on ADM by MTT method, cells apoptosis, apoptotic proteins and related molecules expression after HSP27-specific antisense oligonucleotides insertion that down-regulate HSP27expression.Results1、MTT results revealed that single adriamycin application significantly promoted the Raji cells apoptosis as dose-dependent and time-dependent manner (P<0.05).2、Flow cytometry method results revealed that bicyclol significantly inhibit the Raji cells apoptosis induced by ADM (P<0.05).3、Bicyclol up-regulated the HSP27expression and inhibited apoptotic factors p-JNK, p-c-Jun, Bax and Cytc activation induced by ADM. However, the inhibitive effect of bicyclol to ADM was alleviated as HSP27expression down-regulated. Conclusion1、Adriamycin significantly inhibit the Raji cell proliferation and promoted its apoptosis as dose-dependent manner.2、The Raji cells apoptotic effect of ADM had been inhibited when treated with bicyclol. The mechanism may be that bicyclol up-regulated the expression of HSP27, which inhibited the activation of apoptotic factor, such as p-JNK, p-c-Jun, bax, Cytc, and affected the apoptosis of cells.
