Abstract/Summary:
Objective Detect the expression of CT259 and CT703 encoded by Chlamydia trachomatis(Ct) under persistent infection or acute infection conditions in HeLa cells,which will supply the basis for exploring the significance of CT259 and CT703.Method1.Establishment of the cellular model of Ct persistent infection using IFN-γ.The model was evaluated by Giemsa staining and electron microscopy obeservation.2.Assayed by RT-PCR,the mRNA expression of CT259,CT703 was detected in HeLa cells under Ct persistent infection or acute infection conditions.3.Assayed by Western-blot,the proteins level of CT259,CT703 was facetted in HeLa cells under Ct persistent infection or acute infection conditions.Result1.Establish the model of Chlamydia trachomatis persistent infection2.It was found that CT259 mRNA was expressed in HeLa cells 6 hours later after Chlamydia trachomatis infection both in persistent infection and acute infection.CT703 mRNA was detected 12 hours later after Ct infection.The expression level of CT259,CT703 mRNA under Ct acute infection conditions was significantly lower than under Ct persistent infection conditions at the same time points.3.The results assayed by Western-blot showed that the protein of CT259 and CT703 could be detected in HeLa cells 24 hours later both under Ct acute infection and persistent infection conditions.The expression was time-dependent under the acute infection conditions. The protein levels were significantly lower under the persistent infection conditions compared with that under the acute infection conditions at the same time points.Conclusions1.The model of Chlamydia trachomatis persistent infection was successfully established.2.The expression of CT259 or CT703 mRNA in Chlamydia trachomatis under the persistent infection conditions was significantly less than that of the acute infection conditions in HeLa cells.3.The proteins coded by CT259 and CT703 in Chlamydia trachomatis under the persistent conditions were less than that of the acute infection conditions in HeLa cells.
