OBJECTIVE: To study the expression of PI3K,Akt and ERK1/2 in colorectal carcinoma and the relationship between the expression and the clinicopathological characteristics.METHODS: To select 40 fresh specimens taken from resected colorectal carcinoma and another 40 from the tissue beside the colorectal carcinoma. Tissue sections were stained by EnVisionTM immunohistochemistry with Akt as polyclonal antibody. Tissue sections were stained by immunohistochemical S-P method with PI3KP85 as polyclonal antibody and ERK1/2 monoclonal antibody . The expression of PI3K , Akt and ERK1/2 in colorectal carcinoma and the relationship between the expression and the clinicopathological characteristics were studied with X2 check,t check and the Spearman grade relevance analyse.RESULT: 1.Positive expression of PI3K and Akt in the tissue of colorectal carcinoma were 67.5% and 75.0% , which were obviously higher than those of the tissue beside the colorectal carcinoma (P <0.01). In the colorectal carcinoma,the expression of PI3K and Akt had a positive correlation (r = 0.687, p <0.01). 2,Positive expression of ERK1/2 in the colorectal carcinoma was 57.5% , which was obviously higher than that in the tissue beside the colorectal carcinoma (P <0.01). 3.The expression of PI3K , Akt and ERK1/2 in the colorectal carcinoma were not correlated with the age , tumour location , pathological type , clinical stages, lymph node metastasis and distant metastasis (p>0.05). The expression of PI3K and Akt were not correlated with that of ERK1/2 in colorectal carcinoma (P> 0.05).CONCLUSION: 1, The expression of PI3K and Akt increased obviously in the colorectal carcinoma . In the colorectal carcinoma , a positive correlation was observed between the expression of PI3K and that of Akt,but they had no correlativity with the clinicopathological characteristics. It indicated that the PI3K/Akt signal passway may be an early event of the occurrence and development of the colorectal carcinoma. The abnormal activation of PI3K/Akt signal passway played a certain role in the formation of the colorectal carcinoma.2, The expression of ERK1/2 increased obviously in the colorectal carcinoma . It showed that the expression of ERK1/2 was related to the development of the colorectal carcinoma.
