Objective: Induce mouse embryonic stem cells to differentiate into smooth muscle cells and endothelial cells, now exploring the use of induction of endothelial cells and vascular smooth muscle cells to co-implant on PLGA that has been dialed with gelatin , By constructing small-diameter tissue engineering vascular, detection of embryonic stem cells and PLGA BiocompatibilityMethods:①Induced mouse embryonic stem cells differentiated into endothelial cells (EC) and smooth muscle cells②Detected the contact angle of the PLGA treated with/without 1%gelatin. VSMc was seeded onto PLGA treated and non-treated by gelatin, after 60 minutes with toluidine blue staining to observe the growth of cells③In PLGA-chip plant, respectively, induced by the endothelial cells, smooth muscle cells and embryonic stem cells, divided into A, B, C three groups of training, measured by radioimmunoassay in this group of three culture medium Endothelin(ET)④To mouse embryonic stem cells induced differentiation of endothelial cells and smooth muscle cells in the plant, respectively, within the tubular lumen PLGA surface, build a small-diameter tissue-engineered vascular , and then implant small-diameter tissue-engineered vascular grafts into 4-weeks-old nude mice, at intervals of 1 week sustained 3 weeks out-embedded tissue engineering of small caliber vascular to HE staining, immunohistochemistry and electron microscopy scanningResults:①Induction of smooth muscle cells and endothelial cells consistent with their morphological characteristics②Untreated group contact angle were 87.4°and treated group contact angle were 61.3, Toluidine blue staining showed that smooth muscle cells grown in the material well-distributed③Endothelin levels in cells in A group was 0.81±0.37 pg / ml, B group 5.57±1.23 pg / ml, C group 4.50±0.59 pg / ml④Removed in vivo small-diameter tissue-engineered vascular, by immunohistochemistry, HE staining and scanning electron microscopy show that the lumen of vascular smooth muscle-like cells and endothelial-like cells in the PLGA scaffolds showed growth plate fusion, and see a large number of cells extracellular matrix Conclusion:①The scaffold treated by 1% gelatin improved the cells absorbability②Embryonic stem cells induced by smooth muscle and endothelial cells and tissue engineering materials (PLGA) has good compatibility, PLGA is better used to produce small-caliber vascular tissue engineering scaffold material.
