Objective: To investigate the expression and their correlation of BCRP,HIF-1a and HER-2 in invasive ductal carcinoma of breast,to elucidate their relationships with clinicopathologic Parameters,ER/PR status,and Disease Free Survival Time in human breast cancer. Detecting the expressions of the three proteins before chemotherapy will provide evidence for the selection of drugs and for the judgement of the prognosis.Methods:(1)Expressions and correlations of BCRP,HIF-1a and HER-2 were detected in 75 cases of breast cancer tissue and 15 cases of normal tissue by ElivisionTMplus immunohistochemistry. (2)as well as the correlate with clinicopathologic Parameters,ER/PR status, and Disease Free Survival Time by collecting complete clinical follow-up datas and getting the message about survival condition of the patients,Results: (1) The positive rates of BCRP,HIF-1a and HER-2 expressions were 41.3%(31/75),61.3%(46/75) and 26.7% (20/75) ,respectively. The positive rate of BCRP was 13.3% (2/15) in normal tissue,expressions of HIF-1a and HER-2 were not observed. (2) Expression of BCRP was positive correlated with expression of HIF-1a(r=0.333,p=0.004), and expression of HIF-1a was positive correlated with expression of HER-2(r=0.231 ,p=0.046),but BCRP was not associated with the status of HER-2(r= 0.167,p=0.151). (3) The expression levels of BCRP are not associated with clinicoPathologic Parameters( P>0.05). HIF-la,HER-2 was significantly correlated with the clinical TNM stage and lymph node status(P=0.015,0.021;P=0.006,0.033) ,but had no correlation with age ,tumor size and the status of ER and PR( P>0.05). (4) In 24 patients,the expression of BCRP,HIF-1a and HER-2 was correlated with DFS by Kaplan-Meier survival analysis respectively(P1=0.047,P2=0.001,P3= 0.042 );The coexpression of them was also correlated with DFS by Kaplan-Meier survival analysis(P= 0.034 ).Conclusions: (1) There exist the overexpression of BCRP,HIF-1a and HER-2 in invasive ductal carcinoma of breast. Expression of BCRP was positive correlated with HIF-1a(r=0.333,p=0.004),Expression of HIF-1a was positive correlated with HER-2(r=0.231 ,p=0.046),whereas there was no correlation between BCRP and HER-2(r= 0.167,p=0.151)。(2) In the process of tumor resistance to medicine, BCRP,HIF-1a and HER-2 can coexist. Moreover, there is interaction of them and this may be unfavourable prognostic indicator in the patients of invasive ductal carcinoma of breast with chemical therapy.
