Objective To investigate the effects of wild-type XPD in SMMC-7721 hepatoma cells,its relationship with wild-type p53,cdk7 and c-myc.Methods We transfected stablely wild-type XPD and empty plasmid to human SMM-7721 hepatoma cells,And took human SMM-7721 hepatoma cells as a control group, with the wild-type XPD cells group and empty plasmid cells group as experimental groups,We detected the XPD,p53,cdk7 and c-myc expression changes in three groups of cells by RT-PCR ,Western blot methods, using MTT and FCM methods detected cell proliferation and cell cycle changes.Results . in three groups of cells, the XPD cells Group, XPD, p53 expression was significantly increased, cdk7, c-myc expression was significantly reduced, they had no difference between the other two groups, the XPD gene inhibited hepatoma cells proliferation. hepatoma cells stagnated in the G1 phase of cell cycle.Conclusion The XPD plasmid transfected into hepatoma cells, the XPD inhibited hepatoma cells. It had a synergistic effect on the inhibition of hepatoma cells. At the same time it hindered the cdk7, c-myc to promote the role of hepatoma cells...
