Purpose:To investigate the effect of interventional therapy of simvastatin on diabetic rats,and the changes in bone mineral density (BMD),bone morphology,bone metabolism and bone tissue bone morphogenetic protein-2(BMP-2),as well as serum interleukin-6(IL-6).Method:After one week'adaptive feeding,thirty SPF 4-months-old male SD rats were randomly selected as group A(normal control group) and group B.A is 10 rats,B is 20 rats.The rats of group A inject citrate buffer liquid in abdominal cavity.After the rats of group B has been established the diabetic model by intraperitoneal injection of streptozotocin,it was randomly selected as group B1(diabetic control group) and group B2(simvastatin treatmement diabetic group).Group B1 and B2 both are 10 rats.The rats of group B2 was administered orally simvastatin(5mg.(kg-1·d-1)),as well as group A and group B1 were administered orally 1.5ml normal saline every day.At the end of 14 weeks,the BMD was determined.Then all group rats were killed,bone morphology,serum alkaline phosphatase(ALP),serum tartaric-resistant acid phosphatase(TRACP),and bone tissue BMP-2 gene expresses,as well as serum IL-6 and so on were investigated.Result:BMD:Compare with group A,the group B1 was significantly decreased(P<0.01);Compare with group B1,the group B2 was increased (P<0.05).Bone morphology:the trabecular bone in group B1 has decreased, and compared with group B1,the group B2 was improved.Bone metabolism index:compared with group B1,the serum TRACP in group B2 was significantly decreased(P<0.01),and the serum ALP in group B2 was increased(P<0.05).The level of bone tissue BMP-2 gene expresses: compared with group B1,the group B2 was increased(P<0.05).The serum IL-6:compared with group B1,the group B2 was decreased(P<0.05).Conclusion:Osteoporosis feature was found in the STZ-induced diabetic rats after 14 weeks.The simvastatin could increase the BMD, improve the bone morphology and bone metabolism,and enhance the bone tissue BMP2-mRNA expression,as well as decrease serum IL-6 in diabetic rats.This can provide experimental basis on simvastatin improving and treating the diabetes complicated with osteoporosis.
