Objective: The use of certain parameters of the low-intensity ultrasonic radiation SD rat skin. Observed the low-intensity ultrasound on the biological effects of skin aging. Explored the low-intensity ultrasound treatment of the feasibility of skin aging and proved a theoretical basis for for the treatment of skin aging with the low-intensity ultrasound .Methods: 15 male Sprague-Dawley(SD) rats were randomly divided into four groups: The 1D group were taken biopsies at 1 day after treatment(1D group),. the 7D,14D and 30D groups were 7 day,14 day,30 day after treatment(7D,14D,30D). The control group were set with non-treated areas of each rat(C group). The 15 SD rats were treated at three different regions with the low-intensity ultrasound, in an intensity of 1.0W/CM2,a frequency of 43KHZ. AT day 1,7,14,30 after treatment, skin specimens ,taken from treated and non-treated areas of each rat, were processed for routine paraffin sections and stained with haematoxylin and eosin(H&E) and measured for the expression of typeâ… andâ…¢pro- collagen mRNA and collagen with RT-PCR and immunohistochemistry was also performed.Result: (1)The integrity of skin tissue was identified morphologically and structurally after treatment in microscope. Stains of skin collagens revealed no significant difference between non-treated regions and treated regions at day 1, 3. Collagens stains were stronger in treated regions than those in non-treated areas at day 7.Collagens stains were obviously enhanced at day 30,special in Typeâ… ,â…¢. The dermis was found thickening and the arrangement of collagen was dense and orderly through histology observation. (2) The expression of typeâ… pro-collagen mRNA were stronger in treated regions at day 14. At day 30 after treatment , the expression of typeâ… pro-collagen mRNA significantly increased in treated regions(p<0.05). The expression of typeâ…¢pro-collagen mRNA were stronger in treated regions at day 7. At day 14,30 after treatment , the expression of typeâ…¢pro-collagen mRNA significantly increased in treated regions(p<0.05). (3) The typeâ… andâ…¢collagens stains were stronger in treated regions than those in non-treated areas at day 14.30 days after treatment with significant increase (p<0.01).Conclusion: (1) The expression of typeâ… andâ…¢pro-collagen mRNA significantly increased in treated regions, Collagens stains were obviously enhanced after the low-intensity ultrasound exposure. (2) After treatment, the dermis was found thickening and the arrangement of collagen was dense and orderly. (3) It can be concluded that low-intensity ultrasound radiation could facilitate the restoration of skin elasticity by increasing the content of collagens in skin and might be utilized for the treatment of skin aging. Also the low-intensity ultrasound had the characteristics of non-invasive, no damage, no scar and pigment formation.
