Lignum Dalbergiae Odoriferae is the dry heart wood of trunk and root of Dalbergia odorifera T.Chen which can promote gas circulation to relieve pain and promote blood flow. And volatile oil is the major component of it,which contains nerolidol, bisabolene, farnesol and so on.Xiangdan injection is mixed by Salvia extract and saturated solution of Jiangxiang by steam distillation. It has the function of removing blood stasis, promoting blood circulation, expanding blood vessels and improving the flow of aeteria coronaria. In clinic Xiangdan injection is commonly used for heart disease, coronary artery disease and cerebrovascular disease, and it have good effect in hepatic cirrhosis and chronic renal failure.In the Xiangdan injection process,the amount of jiangxiang and danshen is 1000g to 1000g,and it is the aromatic water of jiangxiang which is used,but the volatile oil on the water is discarded,so a great part of the oil is wasted,this passage questiones the amount of jiangxiang in the prescription, a reasonable amount of jiangxiang should be innovated,and we should research depeatedly and systemly the volatile oil of jiangxiang, which contains mainly as follows:(1)using GC method,to determine the amount of d-nerolidol in volatile oil, carrying on the methodology inspection to the best chromatograph condition, to build the best method for the experiments;(2) using GC method, to determine the amount of d-nerolidol and volatile oil of diffent kinds of jiangxiang raw material,to inspect the quality of them;(3) using GC method, to determine the amount of d-nerolidol in the aromatic water of jiangxiang,the volatile oil saturated solution and the salvia miltiorrhiza solution, Different concentrations of Tween– 80's salvia miltiorrhiza volatile oil saturated solutions to carry on the content determination, inspecting the solubility of jiangxiang volatile oil and the influence of the Different concentrations of Tween–80 and danshen extracts to the volatile oil;(4) Using the GC method, to determine the amount of d-nerolidol of xiangdan injection of different factories and study the quality of them,then provide a reasonable amount of jiangxiang law material for them;(5) Using the GC-MS method, to determine the chemical components of the volatile oil and aromatic water of jiangxiang, and to compare and analyze the difference of them,to provide reasonable quality control application of the volatile oil for the prescription;(6) Carring on the acute toxicity experiment of the volatile oil, the mouse's bearing of the oxygen deficition to determine the volatile oil's smallest activity and it's safety,and unifying the above experiments to determine the jiangxiang raw material's reasonable amount in the prescription,and what's more it can also provide a sciertific reference of disigning , quality control and the reasonable amount in clinical for the injections which contains volatile oil.Following results have been obtained through the above experiments:(1) The GC method,with which to determine the amount of d-nerolidol of volatile oil, has many merits,such as convenience, accurace, stability,and the values are seperately 0.67%,0.85%,1.0%; the concentration of the d-nerolidol is good between 0.15mg/ml-0.42mg/mL,and it's average recovery was 99.90%;(2) the amount of d-nerolidol and the volatile oil of the ten different origin raw material of jiangxiang is 0.07%-1.24% and 40.35%-53.03%.(3) In the 500ml aromatic water from the 50g,100g,250g,500g,1000g jiangxiang raw material seperately, the amount of d-nerolidol is seperately 17.8,17.96,17.58,18.09,17.95mg,which equivalate to 1.54,1.55,1.51,1.55g jiangxiang raw material; Saturated aqueous solution of volatile oil and its danshen solution is seperately 17.51,19.40mg, which equivalate to 1.51,1.67g jiangxiang raw material,but after adding Tween-80 in them, the solubility of the volatile oil increases greatly, in 0.3%,0.5%,0.7% Tween-80 solution, the amount of d-nerolidol is seperately 130.21,179.92,178.43mg, which equivalate to 11.21,15.49,15.36g jiangxiang raw material; in 0.3%,0.5%,0.7% Tween-80 danshen solution, the amount of d-nerolidol is seperately 154.99,182.36,181.70mg mg,which equivalate to13.34,15.70,15.60g jiangxiang raw material;(4) the amount of the d-nerolidol in xiangdan injections of different factories is great different, the amount of d-nerolidol, except two of them ia thero,the others are between 5.87 and 102.18mg,considering the jiangxiang raw materail coming from different areas mentioned in the fourth chapter, the jiangxiang raw material needed in the 10000ml xiangdan injection is 0.47-156g.(5) Both of the aromatic water and the volatile oil of jiangxiang contains the same component of nerolidol and (7R,10S)-2,6,10-Trimethyl-7,10-epoxy-2,11-dodecadien-6-ol.(6) the acute toxicite experiment indacates that the LD50 is 125.84mg/kg,the oxygen deficit experiment and the myocardial ischemic injury experiment indicates that the volatile oil and the aromatic water of jiangxiang can't improve the condition of the mouses'deficiency of oxygen and have no effection on myocardial ischemic injury experiment;(7) By studying the solubility volatile oil in the xiangdan Injection and its impactors the regions where it lives and the manufacturers which make it and the chemical composition of it,and considering the above experiments and the acute toxicity experiment,it comes out that a reasonable amount scope of jiangxiang raw material used in xiangdan injections is from 1.42-146g.
