Objective: To detect protein expression of IGF-1 and TNF-a in coronary artery of patients with coronary atherosclerosis and analyze the relation between IGF-1 and TNF-a with coronary atherosclerosis, approach the relationship between the inflammatory index and plaque stability, to provide the experiment proof for clinical diagnosis, treatment and prognostic evaluation.Method: Considering the medical history and gross observation of 60 autopsy cases, they were classified as 15 normal cases and 45 athersclerosis coronary artery. The samples were divided into three different groups, designated as normal atherosclerosis (15 samples), stable plague groups (27 samples), and unstable plague groups (18 samples). These specimens were detected by immunohistochemistry, locate macrophage (CD68) and smooth muscle cell (α-actin) in the plague. Nonparametric statistics and Spearman test are applied in statistics treatment.Result: There is no expression of IGF-1 and TNF-a in normal group, but the expression increases in coronary artery atherosclerosis and mainly locates in the smooth muscle cells,foam cells,membrane of endothelial cells and macrophages. Statistical analysis demonstrates that there is more expression in unstable plague group than in normal group and stable plague group.This difference is significant (P<0.05).Positive association between IGF-1 and TNF-a atherosclerosis by Spearman test(rs=0.493 ,P=0.001). Furthermore,we find the expression of TNF- a in intima is more significant than in media(P=0.025).Conclusion : (1) IGF-1 and TNF-a expression can be tested in atherosclerosis and expression level is higher in unstable plagues than stable ones, while no expressions exist in control group. This indicates IGF-1 and TNF-a are highly relevant to the unstable plague.(2) Positive association between IGF-1 and TNF-a in the region of atherosclerotic plaque indicates that IGF-1 is in coordination with TNF-a during the development of atherosclerosis.
