Experimental Research On Intervening Depression With Modified Decoction Of Small Bupleurum

Objective:To discusses the mechanism by observing therapeutic action about Modified Small Bupleurum decoction to depressed model rat,which would provide experimental and theory basis for classical prescription in treating and preventing depression.Methods::48 SD rats were randomly divided into six groups:normal group(8),model group (8),Modified Decoction of Small Bupleurum in low-dose group(8),Modified Decoction of Small Bupleurum in the medium -dose group(8),Modified Decoction of Small Bupleurum in high-dose group(8),Western medicine fluoxetine group(8).Later four groups after swimming in the 4℃water for 5 minutes,clip the tail for 5 minutes,no drinking for 24 hours, no eating for 24 hours,day and night reversed,electric shocking the plantar surfaces of toe for 5 minutes(1mA 30V) stimulated by a random arrangement at 30 days,one day species,to made the animal models of depression.Blood and brain frontal lobe were taken by cutting the head,and in accordance with the pathogenesis of TCM,respectively,from the onset of central monoamine neurotransmitter function of the imbalance theory,cell theory of immune factors, abnormal lipid metabolism in the perspective of theory of three mechanisms General observation.Selected high-density lipoprotein,triglyceride,cholesterol,norepinephrine,DA, 5-HT,TNF,IL-1,IL-4,IL-6,such as sensitivity and specificity are high indexes to carry out comparative study of multiple.Result:Weight changes,DA,5 - HT,NE in the brain,IL-1,IL-2,IL-6,TNF,total cholesterol, triglyceride,high density lipoprotein in plasma,such as content as follows:blank control group and model group were significantly different(P＜0.05);the blank group and the fluoxetine group was no significant difference(P＜0.05);the model group and the fluoxetine group were significantly different(P＜0.05);in model group with Modified Decoction of Small Bupleurum in medium & high-dose group were significantly different(P＜0.05);There were no significantly difference in weight and high density lipoprotein during the fluoxetine groupand Small,Medium,Large-dose groups of groups Modified Decoction of Small Bupleururn(P＞0.05).Conclusion:Modified Decoction of Small Bupleurum,especially in medium & high-dose groups,has a good result in treating the depression.
