Objective: To observe the variation of serum total cholesterol(T-CHO), triglyceride(TG),high density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol (LDL-C),plasma semicarbazide-sensitive amine oxidase (SSAO) activity and plasma formaldehyde concentration in the process of rabbit artherosclerosis and investigate possible mechanisms of 2-bromoethylamine improve artherosclerosis.Methods: 30 healthy male rabbits were randomly assigned to four groups, fed with the follow ingredients:â‘ normal rabbit diet (control group,n=6).â‘¡1% cholesterol diet (HC group,n=8).â‘¢1% cholesterol & 0.2% methylamine diet (MA group,n=8).â‘£1% cholesterol & 0.2% methylamine diet & ear edge vein inject 2-bromoethylamine (100mg/kg,iv)on the 9,the 10,the 11 week end(2-BEA group,n=8 ).The rabbits were fed with these diets respectively for 12 weeks. Blood samples were collected from animals ear central artery for Serum T-CHO,TG,HDL-C,LDL-C,plasma SSAO activity and plasma formaldehyde measurement on the 0-week origin,the 9-week end and the 12-week end respectively.At the end of experiment, endothelium-dependent vasorelaxation of the isolated rabbit thoracic aorta was assayed and pathological morphology was observed by HE staining,oil red O staining and Sudanâ…£staining.Results:1. The levels of T-CHO ,HDL-C and LDL-C of the HC group were significantly higher than the control group (P<0.05) on the 9-week end and the 12-week end; the level of T-CHO in the MA group were significantly lower on the 12-week end than that on the 9-week end (P<0.05), the level of LDL-C in the 2-BEA group increased significantly on the 12-week end than that on the 9-week end (P<0. 05).2. Compared with the control group, the plasma SSAO activity and plasma formaldehyde concentration of the HC group both increased significantly (P<0.05) on the 9-week end and the 12-week end; the plasma SSAO activity of the 2-BEA group decreased significantly on the 12-week end than that on the 9-week end (P<0.05) and the plasma formaldehyde concentration increased slowly; the plasma SSAO activity of MA group had no significance difference and the plasma formaldehyde concentration significantly increased on the 12-week end than that on the 9-week end (P<0.05).3. Endothelium-dependent vasorelaxation to acetylcholine were significantly impaired in the aortas of HC and MA groups (P<0.01). The blood vessels endothelium-dependent dilation improved significantly in the 2-BEA group than the MA group (P<0.01).4. The pathological morphology figures indicated a reduction in plaque area in aortas of the 2-BEA group, compared with the MA group;there were no significant differences in plaque areas in aortas between the HC and MA groups.Conclusions:1. With rabbit artherosclerosis aggravating, plasma SSAO activity gradually increased and plasma formaldehyde concentration also elevated in the process of artherosclerosis.2. 2-bromoethylamine could improve endothelium dependent vasodilatation and reduce atherosclerotic plaque area, meanwhile could inhibit increasing of SSAO activity and formaldehyde concentration in the process of rabbit artherosclerosis.
