Expression And Significance Of IGF-1,MMP-2,MCP-1 Protein In Different Gastric Tissues

Objectives:We have aimed to study the expression of IGF-1,MMP-2,MCP-1protein in different gastric tissues and to evaluate the significant correlation between their expressions and cell cycle,to investigate their possible roles in the gastric carcinogenic mechanism,to provide evidence for prevention and therapy of gastric carcinoma as a therapeutic target.Methods:Gastric tissue samples were obtained from 20cases with chronic superficial gastritis(CSG),35 cases with intestinal metaplasia (IM),30cases with dysplasia (Dys) and 56 cases with gastric carcinoma (GC). Expressions of IGF-1,MMP-2,MCP-1 protein were dectected by immuno -histochemistry(S-P method)and western-blot and cell cycle by flow cytometer(FCM).The datas were analyzed by software SPSS 13.0, chi-square test was used to compare the difference between groups.A difference was considered significance if the p value was below 0.05.Results: Expressions of IGF-1,MMP-2,MCP-1showed a gradually increasing tendency from CSG, IM, Dys to GC. Positive expression rates of IGF-1,MMP-2,MCP-1were 15.0%,10.0% and 5.0% in CSG,37.1%,20.0% and 11.4% in IM,56.7%,20.0% and 20.0% in Dys,87.5%,83.9% and 89.3% in GC respectively. There was a significant difference between either two of the four tissues in expression of the IGF-1 proteins(P<0.05);There was a significant difference between GS and CSG,IM,Dys in expression of the MMP-2,MCP-1 proteins(P<0.05),but no significant difference were found compared CSG,IM and Dys with each other.(P>0.05).In histological, significant relations of the expression of IGF-1andMCP-1 were found in TNM,lymph nodes metastasis and histological type(P<0.05); significant relations of the expression of MMP-2were found in TNM,lymph nodes metastasis(P<0.05);but no significant relations were found in its histological type. Also no significant relations were found assocated with their years,sexes,diameters of tumors and so on(P>0.05).Western Blot result:CSG,IM,Dys,GC tissues display to comprise three idio-straps,their relative molecular weight are7.6KD,15KD,72KD,and precancerous change tissues display weaker than the carcinoma tissues。Growth-rate showed a gradually increasing tendency in gastric carcinoma from fast-growth to no-growth,positively correlated with the expression of IGF-1protein in different gsastric tissues .Conclusion: Expression of IGF-1,MMP-2,MCP-1showed a gradually increasing tendency in gastric carcinogensis,they may promote gastric carcinoma. Also they promote cell proliferation. Expression levels of IGF-1,MMP-2,MCP-1can be regarded as a marker for evaluating malignant degree of GC, lymph node metastasis and prognosis.
