To observe the therapeutic effect of Rukuaixiao Decoction on hyperplasia of mammary gland (MGH) in rats and to reveal the mechanism, providing the basis for the wider clinical use.90 unpregnant SD female rats were radomly divided into control group (C), model group (M), tamoxifen group (TAM), different dose of Rukuaixiao groups (RKX) and different dose of Radix salviae miltiorrhizae groups (SM). Injection of estradiol and progesterone were given to establish rat models of mammary gland hyperplasia. The rats were administrated with 2mL normal saline, 2mL normal saline, 2mL tamoxifen, 5.8g/kg, 11.6g/kg and 23.2g/kg Rukuaixiao decoction, 1.2g/kg, 2.4g/kg and 4.8g/kg Radix salviae miltiorrhizae respectively one time a day. The rats were euthanatized after 30d. The breast diameters, teat heights, visceral coefficients and levels of serum sex hormones were measured. The blood rheology, SOD activity and MDA contents in liver and serum were also estimated. Pathologic changes of mammary gland in rats were observed by microscopy. The results show as follows:1. Injecting rats with estradiol and progesterone induced the model of Mammary Gland Hyperplasia (MGH). The results show as follows: Compared to the control group, breast diameters and teat heights increased significantly and body weight and thymus index decreased much more on rats in model group (P<0.01). The number of mammary lobule was increased. Lumen of catheter and alveolar cavity were obvioulsly enlarged on rats in model group. Serum E2 and PRL in model group increased higher than that of the control group (P<0.01). P in model group were lower than that in control group (P<0.05).2. Different doses of Rukuaixiao decoction could significantly decrease the diameters and heights of second pairs of teats, reduce the numbers of mammary gland lobules and secretion, and relieve the pathologic changes of mammary gland. Radix salviae miltiorrhizae did not affect the pathologic changes of mammary gland.3. Rukuaixiao decoction could reduce the contents of serum E2 and PRL, and increase the serum P content. The results suggested that RKX could regulate endocrine dyscrasia. 4. Rukuaixiao decoction could decrease blood viscosity, plasma viscosity, hematocrit and red blood cell rigidity index. Radix salviae miltiorrhizae could also obviously improve the hemorrheological characteristics of rats of MGH.5. All Rukuaixiao groups could increase the activities of SOD in liver and serum, especially middle dose group of RKX. RKX could also reduce the contents of MDA in serum. Radix salviae miltiorrhizae could obviously improve the activitiy of SOD in serum and reduced the contents of MDA in liver and serum.
