Objectives To observe the relations of HPA-1-6,15 alleles and idiopathic thrombocytopenic purpura(ITP), and study the expression levels of toll like-receptor-2,4,6,7,9 in peripheral blood mononuclear cells (PBMCs) from ITP patients.Methods Polymerase chain reaction-specific sequence primers (PCR-SSP) method was used to genetyping HPA-1-6,15 in the ITP group (n=25) and control of healthy group (n=25). The platelet adsorbed and free antibodies were detected in two groups by ELISA and microcolumn gel immuassay. We performed fluorescence quantitative RT-PCR analyses of PBMCs from the ITP group (n=30) and control of healthy group (n=30),to estimate TLR2,TLR4,TLR6,TLR7 and TLR9 mRNA expression levels.Results 1)The allele frequencies of HPA-3 was a statistically significant difference between the ITP group and control group (P<0.01),and the frequencies of HPA-3a in ITP group was higher than in control group, and the frequencies of HPA-3b was lower than in the control group ,the frequences of other alleles was no significant difference in two groups(P>0.05); 2)The platelet free antibodies were detected in 17 ITP samples(68%) and the adsorbed antibodies were detected in 21 ITP samples(84%), the platelet antibodies were no detected in the control group; 3)The relative TLR2 and TLR7expression levels of PBMCs from the ITP group were significantly higher than those in control group, and the relative expression levels of other TLRs was no significant difference in two groups(P>0.05).Conclusion 1)This study suggests there was an probable association between the HPA-3 allele and ITP; 2)The detectable rate of platelet adsorbed antibodies significant higher than platelet free antibodies;3)Up-regulated expression of relative TLR2 and TLR7 mRNA levels in PBMCs from patients with ITP, and probablely play an imprtant role in the pathogenesis of ITP.
