Objective To evaluate the significance of tissue angiotensinⅡ(AngⅡ) and vascular smooth muscle cell between proximal and distal sites of abdominal aortic stenosis in atherosclerotic rabbits.Methods We divided New Zealand rabbits into common diet group and high-lipid diet group.After 28 weeks, we referred to them different operations, and divided them into 4 groups, high-lipid operation group(ho group),high-lipid sham operation group(hso group),common operation group(co group) and common sham operation group(cso group). After operations, we continued to keep them 4 weeks, measured their blood TG,TC,LDL and measured their blood pressure in different positions of abdominal aortas, took their abdominal aortas, HE-staining was used to identify the thickness of intima and media, Anti-α_action and AngⅡantibodies were used to identifyα-actin and AngⅡwith immunohistochemistry.Results1. TC(23.03±2.62 VS 1.95±0.14),TG(4.77±0.81 VS 1.48±0.27),LDL(10.73±1.12 VS 1.75±0.56)of high-lipid diet group were more than commom diet group,p<0.05;2. Blood pressure (BP) at the proximal sites of constriction was higher than that in distal sites in both ho group(84.18±5.28 VS 54.98±3.59) and co group(72.30±13.41 VS 54.11±11.00), p<0.01; 3.Media thickness(MT) at the proximal sites of constriction was higher than that in distal sites in both ho group(174.42±7.83 VS 128.35±8.13) and hso group(154.04±3.22 VS 136.13±4.30), MT at the proximal sites of constriction in ho group was higher than that in hso group, p<0.01;4.Intima thickness(IT) at the proximal sites of constriction was higher than that in distal sites in ho group(183.76±10.53 VS 118.63±13.35), p<0.01;IT at the proximal sites of constriction in ho group was higher than that in the same sites in hso group(183.76±10.53 VS 164.59±4.96), p<0.01;5. The level ofα-actionIn intima, The level ofα-action at the proximal sites of constriction was higher than that in distal sites in ho group(0.13±0.01 VS 0.08±0.01), p<0.01; The level ofα-action at the proximal sites of constriction in ho group was higher than that in the same sites in hso group(0.13±0.01 VS 0.10±0.02), p<0.01;In media, The level ofα-action at the proximal sites of constriction was higher than that in distal sites in both ho group and co group, but in ho group(0.23±0.04 VS 0.15±0.02)p<0.05,in co group(0.10±0.03 VS 0.08±0.03)p>0.05; The level ofα-action at the proximal sites of constriction in ho group was higher than that in the same sites in both hso group and co group, p<0.05;6.The level of AngⅡ:In intima, The level of AngⅡat the proximal sites of constriction was higher than that in distal sites in ho group(0.26±0.01 VS 0.12±0.01), p<0.01; The level of AngⅡat the proximal sites of constriction in ho group was higher than that in the same sites in hso group(0.26±0.01 VS 0.17±0.01), p<0.01; In media, The level of AngⅡat the proximal sites of constriction was higher than that in distal sites in both ho group and co group, but in ho group(0.28±0.04 VS 0.16±0.02),p<0.05,in co group(0.11±0.01 VS 0.08±0.02),p>0.05; The level of AngⅡat the proximal sites of constriction in ho group was higher than that in the same sites in both hso group and co group, p<0.05;Conclusions In AS rabbits, BP elevation alone induced by abdominal aortic stenosis was not able to engender media thickness and AngⅡincrease,only the effect interaction between high BP and high cholesterol caused media thickness associated with increases inαactin and tissue AngⅡ.
