OBJECTIVE: This study investigated the effects of a combination therapy of marrowstromal cells (MSCs) and statins (atorvastatin) on myocardial infarction in rats.METHODS:①Harvest mesenchymal stem cells from femur and tibia of six weeks old SD rats that were cultivated. Labeling of MSCs with 5-bromodeoxyuridine (BrdU). To detect the MSCs after transplantation, cells were labeled with BrdU in vitro. Cell surface marker.②Establish acute myocardial infarction model. Ligate the left anterior descending coronary artery .The rats that modeled successfully were divided into 4 groups: control group; atorvastatin group;MSCs group; combination group.③four weeks later, leftventricular systolic pressure, end-diastolic pressure, the maxim alincrease and decrease rate of left ventricular pressure (±dp/dtmax) were determined with physiological recording device. Cytohistology change was detected with immunohistochemical method.RESULTS: Totally 40 successfully modeled rats were involved in the result analysis.①Cell surface markers showed negative for CD45 and CD31, while the CD44 and CD54 were strongly positive. hemodynamic determination: Functional improvement was seen in animals treated with MSCs or atorvastatin alone. However, functional improvement was seen in animals receiving combination therapy.②detection of heart histology: microscopic analysis showed that significantly more MSCs were present in animals receiving combination therapy than in those receiving MSCs alone.③blood vessel density: more new blood vessels were present in animals receiving combination therapy than in other groups.CONCLUSION: The combination of MSCs and atorvastatin for acute Myocardial infarction can promote the therapeutic effects cooperatively.The therapeutic effect of the combination is superior to that of MSCs transplantation or atorvastatin alone.
