Objective:Erectile Dysfunction(erectile dysfunction,ED) a higher rate of incidence of its men and their partners have a significant impact on the quality of life.With the aging population in the community share of the total population of more and more large,the number of older ED patients is increasing.The United States,Massachusetts male aging study(MMAS) report that at a random sample of 40-70 year-old respondents in the prevalence rate of ED up to 52%.Recent study found that certain diseases are defects in ion channel gene mutations and functional results,known as ion channel disease. Although many diseases do not belong to ion channel disease,but the disease occurred in a particular aspect or phase with the ion channel dysfunction is closely related to,but also found that many drug treatment and prevention of diseases through the role of ion channels in the effect.Therefore,the physiology and disease and study on the relationship between ion channel has gradually become one of the hot current research.Corpus cavernosum smooth muscle relaxation or contraction of the regulation,that is cavernosum smooth muscle physiology of the nervous decide separately penis erection or weak status.Evidence suggests many other vascular tissue,the potassium channel, calcium channel corpus cavernosum smooth muscle tension are the main regulators.In cultured corpus cavernosum smooth muscle cells and free corpus cavemosum smooth muscle strips studies have shown:the participation of erectile neurotransmitters mostly smooth muscle cells through ion channels and transmembrane ion flow and the role of nervous regulation of corpus cavemosum smooth muscle.The present study are more Ca2+channel.Study shows that,L-type Ca2+channel Cav1.3 transhipment at Ca2+played an important role,Ca2+channel receptor,ryanodine receptor(RyR) of three subtypes of RyR1,RyR2,RyR3 in the regulation of Ca2+also play an important role,the subject of experimental study of animal L-type calcium channel(CaV1.3) and ryanodine receptor RYR1 in aged rats rat corpus cavernosum penis of expression,for exploring aging and erectile function obstacles(ED) provide a basis for the mechanism.Methods:A group (2-month-old) and B group(18-month-old) male SD rats of the 10 measured serum testosterone,using RT-PCR and immunohistochemical analysis of the two groups at the penis CaV1.3 and RyrR1 The expression of corpus cavemosum.Results:A group of the average value of serum testosterone(T) for 274.426±96.371ng/dl,B group was 135.642±42.431ng/dl,B group than A group of serum testosterone decreased significantly(P<0.05). Immunohistochemical data using integral optical density(IOD) for semi-quantitative analysis:B group CaV1.3 expression(IOD=18.65±8.47) than the A group(IOD=75.48±14.28) significantly reduced(P<0.05);B group RYR1 expression(IOD=21.37+9.64) than the A group(IOD=78.23±13.57) significantly reduced(P<0.05).CaV1.3 and RyR1mRNA two groups at the expression of rat corpus cavernosum using average gray value:B group CaV1.3mRNA expression(mean gray value=0.382±0.046) than the A group (average gray value=1.137±0.415) were significantly lower(P<0.05);B group RyR1mRNA expression(mean gray value=0.146±0.053) than the A group(average gray value=1.215±0.261) significantly lower(P<0.05). Conclusion:Expressed reduce of CaV1.3 and Ryrs1 is one of the mechanisms of aging-ED in aging rat corpus cavernosum.
