Objectives:To observe the effect of yikan capsules on phenotypic transformation of corpus cavernosum smooth muscle cells (CCSMC) and ultra-structure in rats with erectile dysfunction(ED). Methods:50rats were randomly divided equally into5groups, in which10rats were assigned in normal group,40rats were made into erectile dysfunction models by ligating bilateral internal iliac arteries. After ligation,10rats were assigned in control group, and the other30rats were given yikan capsules with doses of1.88g/kg,0.94g/kg,0.47g/kg, respectively. After one month, each rat was injected apomorphine subcutaneously to record the times of yawning and penile erection. Afterwards, expressions of calponin-1and osteopontin(OPN) in CCSMC were examined via immunohistochemical staining, and ultra-structures of CCSMC were observed with transmission electron microscope. Results:After injection of apomorphine, times of penile erection of rats in control group decreased, compared with normal group, suggesting significant difference between two groups. Times of penile erection of rats given yikan capsules increased, compared with control group. Results of immunestochemical staining showed that expressions of calponin-1were decreased, while, expressions of OPN were increased in CCSMC in control group. Whereas, when given yikan capsules, expressions of calponin-1were increased, and expressions of OPN were decreased in CCSMC. Examination with transmission electron microscope showed that there was severe swelling in mitochondrion, mitochondrial crista decreased, or disappeared, even vacuolated in control group. The swelling in mitochondrion has been improved in rats given yikan capsules. Conclusions:It is shown that yikan capsules could improve penile erection of rats with ED, inhibit phenotypic transformation of CCSMC from contractile to synthetic type, and improve morphological structure of mitochondrion in CCSMC. |
