Objective:To observer the curative effect of CPT-11(Irinotecan) chrono-chemotherapy on the nasopharyngeal carcinoma in nude mice,the toxic and side effects of CPT-11(Irinotecan) chrono-chemotherapy on the nude mice with the nasopharyngeal carcinoma,and the cell cycle change of nasopharyngeal carcinoma in nude mice induced by CPT-11(Irinotecan) chrono-chemotherapy.Methods:Fifty nude mice with nasopharyngeal carcinoma were maintained for 3 weeks on a normal chow diet and housed in a room with a 12:12-h light-dark cycle and an ambient temperature of 22℃. These mice were divided into two groups randomly:the mice in control group were injected intraperitoneally with normal saline at 3,9,15,21 HALO(Hours After Light Onset) for 3 days and the mice in experimental group were injected intraperitoneally with CPT- 11 66mg/kg for 3 days at 3,9,15,21 HALO (Hours After Light Onset).The mice were killed at 15 day after the first injection.The blood were taken and WBC,HB,PLT of each mouse were detected.Then tumor xenografts were removed and the inhibitory rates were exanimated.The cancer cells suspension were prepared for detection of cell cycle change by flow cytometry.The change of intestinal mucosa tissue was observed with HE staining on light microscope.Results:CPT-11 shown the significant therapeutic effect on nasopharyngeal carcinoma in the mice of experimental group compared with the mice of the control group(p<0.01).The largest inhibitory rate of CPT-11 on the carcinoma was in 15 HALO(p<0.05) and the smallest was in 21 HALO(p<0.05).In 15 HALO,the WBC, HB,PLT of mice of the experimental group were higher(p<0.05),the body weight was increased more obviously and the grade of the intestinal mucosa damage was lower obviously(p<0.05) than the mice in other experimental groups.In 21 HALO,the WBC,HB,PLT of mice of the experimental group were lowest(p<0.05),the body weight was decreased more obviously and the grade of the intestinal mucosa damage was higher obviously(p<0.05) These in 9HALO were better than in 3HALO(p<0.05).The carcinoma cells of 15 HALO and 9 HALO mainly distributed in the G1 phase(44.2200±19.6499,41.4000±19.70426) and S phase(39.620±13.959,30.0600±23.41582).The carcinoma cells of 21 HALO and 3 HALO mainly distributed in the G2 phase(54.1100±17.33740,53.0300±20.7931).The carcinoma cells of 15 HALO and 21 HALO had significant deviation(P<0.01)。Conclusion: CPT-11 could inhibit the growth of nasopharyngeal carcinoma in nude mice significantly.The curative effect CPT-11 on the carcinoma,the toxic and side effects CPT-11 on the mice with the carcinoma,and the cell cycle changes in carcinoma cells induced by CPT-11 shown the difference in different HALO (15HALO best,21HALO worse?).The results provide a reference for the clinical application of CPT-11 chrono-chemotherapy.
