Objective: To investigate the effect of extract of ginkgo biloba (EGB) on myocyte apoptosisand the expression of associated genes in rat hearts with acute myocardial ischemia and reperfusion.Methods: We choosed inbred,clean wistar rats,their age was 8 weeks,and their weightwas controlled between 250 and 300 gram.30 wistar rats were divided randomly into extract ofginkgo biloba group (EGB group),sham operation group (sham group) and ischemia and reperfusiongroup(IR group).Ischemia-reperfusion model was induced by the left anterior descendingcoronary artery(LAD)ligation for 30 min,followed by 2h of reperfusion.Myocyte apoptosis andthe protein expression of Bcl-2,Bax and P53 genes were detected by terminal deoxynucleotidyltransferase mediated dUTP nick end labeling (TUNEL)and immunohistochemistry .Results: 1.TUNEL results show that:Campared with sham group,the apoptotic indexeswere signifiantly increased in IR group and EGB group(P<0.01) .Compared with IR group ,EGBsignificantly inhibit myocyte apoptosis(P<0.01).2.Immunohistochemical results show that: Camparedwith sham group,the positive expression of Bcl-2 and Bax genes were significantly increasedin IR group and EGB group(P<0.01);Compared with IR group,EGB significantly inhibitedthe protein expression of Bax gene,but increased the protein expression of Bcl-2 geneP<0.01),and raised the ratio of Bcl-2 to Bax incidentally. Compared with EGB group and shamgroup,the positive expression of P53 genes was significantly increased in IR group(P<0.01),butthere was no significant difference between EGB group and sham group(P>0.05).Conclusion:Extract of ginkgo biloba could downregulate the positive expression of Baxand P53 genes and upregulate the positive expression of Bcl-2 gene,thus protect against cardiomyocyteapoptosis from myocardial ischemic reperfusion injury.
