ObjectiveRenovascilar hypertensive rat model was used to detect the expression of ROS and some inflammatory factors TNF-α,IL-6 in the rat myocardial cells.Explored the role of these genes in the development of myocardial hypertrophy of the hypertensive rat may contribute to provide the theoretical and experimental basis for the prevention and treatment of the hypertensive myocardial hypertrophy.Material and Methods1.Renovascilar hypertensive rat model was induced by "Two-kidney One-clip" method.Selected the success models as the experimental group.Rats without any treatment to the renal artery as the controls were operated as well as the experimental group after anaesthetic administration.2.Two dimensional doppler ultrasonography was used to analysis the corresponding index of the hypertensive rat heart and the LVW was determined.3.The expression of ROS in the heart tissue was measured by laser scanning confocal fluorescence microscope.4 The inflammatory factors TNF-α,IL-6 were determined by ELISA Kits.Results1.The expression of the ROS,TNF-α,IL-6 of the blood plasma in each experimental group were significant higher than controls(P<0.05).2.The correlation analysis revealed that the expression of the ROS, TNF-αand IL-6 in blood plasma after modeling positively correlated with that of ROS(r=0.916,P<0.05),TNF-α(r=0.854,P<0.05)and IL-6(r=0.778,P<0.05). ConclutionThe ROS,TNF-αand IL-6 may play important roles in the development of renovascilar hypertensive myocardial hypertrophy.
