ObjectiveFirstly, we setted up the determination methods about paraquat and methomyl in blood. Secondly, we putted paraquat and methomyl standard solution into the health human blood, made it through several kinds of HP adsorption columns, then the blood samples were extracted with ethyl acetate and the rudimental poison was determined by HPLC, and then compared the clearance percentage of poisoning used different absorptions or different time (dosage) in the same type of adsorbent, to find out better adsorbent to use in HP.MethodWe put paraquat or methomyl standard solution into the health human blood, and took it through a MMAC and HA230 resin hemoperfusion cartridge having the huge absorption capacity Then the blood samples were extracted by perchlorate (paraquat) or acetic ether(methomyl), at last the contents of methomyl or paraquat were determined by high performance liquid chromatography(HPLC).ResultWhen the amount of membrane activated charcoal and HA230 adsorbent resin used the Conditions of we choosed, The limit line of paraquat in mankind blood was 0.05ng. The recoveries of low, middle and high three kinds added were between 90.76% and 95.80%. The relative standard deviations (RSD) was between 1.69% and 7.98%. The cleanup-rate was 97.69% -99.52% and 97.90%-98.61%, Membrane activated charcoal has higher percentage than HA230 resin in eliminating paraquat of high doses., the cleanup-rate of low doses between MMAC and HA230 resin have no statistic difference;The limit line of methomyl in mankind blood was 0.02ng. The recoveries of low, middle and high three kinds added were between 96.31% and 99.68%. The relative standard deviations (RSD) was between 2.65% and 4.92%. The cleanup-rate was 98.68% -99.64% and 98.33% -99.30%.the cleanup-rate of all doses between MMAC and HA230 resin have no statistic difference.ConclusionParaquat and methomyl in the blood can be determined by the HPLC and the method is rapid and convenient, meanwhile the result turned out to be reliable. HP is an effective means to eliminate the major part of paraquat and methomyl from the blood. Different sorbents vary in ability of removing different pesticides from blood. the suitable time for hemoperfusionas controlled at 2.0h left and right; the amount of adsorbent was controlled at 1.5g (MMAC) and 0.35g (HA230 resin) of paraquat,, the suitable time for hemoperfusionas controlled at 2.0h left and right; the amount of adsorbent was controlled at 0.5g between the MMAC and HA230 resin of methomyl; In brief, HP can clear away paraquat and methomyl rapidly and effectively in blood. So it's a good therapy to poisoning patients.
