Objective:To explore the effect of YXL granule formulated by the main principle of dissolving blood stasis and unblocking meridian on the Ca2+ -CaM-CaMKⅡαSignal in the hippocampus of Epileptic Kindling Model in Rats induced by PTZ.Methods:9 SD rats were selected randomly from 70 experimental rats as a normal control group.The rest were injected abdominally by PTZ 35mg/kg for 28 days to establish chronic-kindling models,then 36 from built-up model rats were divided randomly completely into four groups:model,YXL,VPA,NIM.After drug-intervertion,epileptic rat's behaviour and EEG were observed,and the Ca2+ concentration of hippocampal neurons synaptosomes were evaluated by Fluo-2/AM and the expression of CaMmRNA,CaMKⅡαmRNA were also tested by RT-PCR of the real time fluorescence half quantify-fixed in rats hippocampus. Results:After 28 days of YXL granule treatment:1.The potential time were prolonged and last-time of epilepsy-attacks were shorten,and the abnormal frequency and amplitudes of EEG waves in epileptic rats' brain were regulated effectively,there was significant diffirence compared between the YXL group and the model group(P<0.05,P<0.01);2.the[Ca2+]i level of synaptosomes in hippocampus of YXL group was lower apparently than that in the model group (P<0.05,P<0.01);3.the reduced expression of CaMmRNA and the improved expression of CaMKⅡαmRNA in rats hippocampus was found obviously,the comparison between the YXL group and the model group showed significant difference(P<0.05).Conclusions:One of the mechanisms about YXL anti-epilepsy effection is to adjust the balance-misadjustment of the Ca2+ Signal in the epilepsia-rats hippocampus.
