Gastric cancer is one of common and malignant tumors.It is reported that the death rate has occupying first or second in malignant tumors,but invasion and metastasis of gastric cancer are main death causes.Therefore,to study and research mechanism of invasion and metastasis of gastric cancer has become the gastric cancer investigative'hot spots.We have found in our study that TGF-β1 plays important roles in the process of gastric cancer's invasion and metastasis.We used SELDI-TOF-MS technology to contrastly analyse the differences of secrete protein expression profiles in culture fluid in vitro of gastric cancer cells(BGC823,MKN45,SGC7901) after treatment of TGF-β1 to explore the mechanism of gastric cancer's invasion and metastasis.At present,there has few reports about using SELDI-TOF-MS to analyse secrete protein stimulated by TGF-β1 at home and abroad.Purpose:SELDI-TOF-MS technology was used to contrastly analyse the changes of protein expression profiles of gastric cancer cells after TGF-β1 stimulates so as to lay theory and experiment foundations for screening useful difference proteins.Method:BGC823,MKN45,SGC7901 used as research materials.1.Serial subcultivation gastric cancer cells BGC823,MKN45, SGC7901 were cultured in vitro to analyse the changes of protein expression profiles of culture fluid.2.The serial subcultivation gastric cancer cells(BGC823,MKN45, SGC7901) were grouped into test and control groups.TGF-β1 were added to the former,otherwise,not in the latter.Cell Culture fluid were collected and centrifuged after 24h,crossing with WCX2 protein chip to detect protein differences.Result:1.We obtained the secrete protein expression profiles of BGC823, MKN45,SGC7901 in WCX2 chip:①We have found thirteen difference proteins in BGC823 after TGF-β1 stimulating,whose M/Z are M4294,M4932,M4945,M4972, M4991,M5015,5036,M5060,M5153,M5180,M5197,M8577,M8784, respectively.②We have found eighteen difference proteins in MKN45 after TGF-β1 stimulating,whose M/Z are M4292,M4931,M4945,M4972, M4990,M5014,M5152,M5178,M7055,M8190,M8570,M8652, M8670,M8780,M9963,M10098,M10523,M11653,respectively.③We have found eight difference proteins in SGC7901 after TGF-β1 stimulating,whose M/Z are M4945,M4972,M4992,M5015, M5180,M7056,M8573,M8604,respectively。2.Comparing three protein eapression profiles of gastric cancer cells after TGF-β1 treatment,we found two useful and common difference proteins,whose M/Z are M4945,M4972,respectively.3.The stable difference expressions of protein secreted in by BGC823,MKN45,SGC7901's what implicated the differences in proteinum quantity and protenium decoration in secrete protein of different diffentiation gastric cancer cells.Conclusion:The biological markers whose the M/Z is M4945,M4972 of gastric cancer characteristic associated with TGF-β1 were screened,which were useful for early prediction and clinical diagnosis of metastasis and recurring of gastric cancer.
