Study On Srum Protein Fingerprint Associated With Metastasis In Postoperative Gastric Carcinoma Patients

Object:To screen the serum biomarkers related to metastasis and develop diagnostic and predictive patterns in postoperative gastic carcinoma patients,then evaluate its clinical significance.Methods:SELDI-TOF-MS and CM10 proteinchip was used to detect the serum protein fingerprint of early postoperative serum from patients with gastric cancer and 26 healthy people retrospectively.After 2 years follow-up,all the patients were divided into 2 groups:metastatic group and non-metastatic group according to the clinical outcome(metastasize or not).And further more,we detected the post-metastasis serum fingerprint in metastatic-group.The detected data were analyzed by Biomarker Wizard Software and the predictive patterns were developed by Biomarker Pattern Software.Results:(1)22 cases of 60 cases postoperative gastric cancer patients developed metastasis after 2 years follow-up.Compared with healthy people group,there were 74 and 69 significant differential protein peaks in non-metastatic group and metastatic group respectively.14 protein peaks were found between metastatic and non-metastatic group.And the predictive pattern(Patternâ… )composed by 2 proteins(M/Z values were 4768 and 8841,respectively)could classify the 2 groups correctly.In learn model,the sensitivity and specificity was 95.46%(21/22) and 86.84%(33/38),the accuracy was 90%(54/60).In test model,the sensitivity and specificity was 81.82%(18/22)and 84.21%(32/38),the accuracy was 83.33%(50/60).(2)Compared with healthy people group,there were both 70 significant differential protein peaks in pre-and post-metastasis group.40 differential protein peaks were found between preand post-metastasis group.And the diagnostic pattern(Patternâ…¡)composed by 3 proteins(M/Z values were 2640,4115 and 13663,respectively)could classify the 2 groups correctly.In learn model,the sensitivity and specificity was 89.47%(17/19)and 94.74%(18/19),the accuracy was 92.11%(35/38).In test model,the sensitivity and specificity was 84.21%(16/19)and 89.47% (17/19),the accuracy was 86.84%(33/38).(3)M/Z 4115,4350 and 4652 were differential expressed among non-metastasis group, (pre-)metastasis and post-metastasis group.Conclusions:SELDI-TOF-MS could be utilized to analyze protein profiling in screening metastatic-related biomarkers and developing diagnostic and predictive patterns,and the developed patterns may be used to predictive the metastasis and/or recurrence in postoperative gastric cancer patients.
